| Objective:The purpose of this experiment is by observing the effect of moxibustion guan yuan, shenshu for the morphological structure of kidney and adrenal gland with kidney-Yang deficiency syndrome rat model and the effect on hypothalamus CRH levels, to explore possible mechanism which may exist about moxibustion on the improvement of the kidney Yang deficiency syndrome.Methods:40SD male rats were randomly and equally divided into4groups according to the table of random number method:black group,model control group, model group,moxibustion treatment group, each group of10.Black group was to be give normal feeding,without any intervention;The model control group with0.3ml of0.9%N.S daily intramuscular injection,for28days; The model group and the treatment group treated with Hydrocortisone Injection according to2.5mg/100g dose14consecutive days to prepare kidney yang deficiency model,after the success of the building, the two groups continue to give a dose of hydrocortisone injection in order to maintain the kidney Yang deficiency syndrome, Moxibustion group was to be give acupoints moxibustion treatment on guan yuan, shenshu,Other groups rats also grabbed.To be put to death after14days treatment, calculated the viscera index and observed the morphological structure changes of kidney and adrenal gland tissue under microscope.And to observe the change of content of CRH in hypothalamus through using Ria method.Result:1. After modeling14days, model group and moxibustion group rats appeared emaciated, Camponotus curled up, reduced locomotor activity, increased urine output, scrotum wet swelling and body weight of the rats of model group and moxibustion rise less than control group significantly (P<0.05), model group and moxibustion group rectal temperature significantly lower than the control group (P<0.05), which can be used to illustrate the model to replicate the success.2. It was shown that the morphological structure of adrenal gland and kidney of rats in model group had obvious glomerular damage, sparse distribution, renal tubular cell wall thinning, renal distal tubule expansion, proximal tubule atrophy of the adrenal cortex; irregular arrangement, unclear structure, thin cortex, medulla slightly thickened. By contrast to the model group,the morphological structure of kidney and adrenal gland in the moxibustion group, there was a certain degree of reduction. 3. After14days of treatment, compared with model group, body weight of rats in the moxibustion group growth rate increased significantly (P<0.05), but less than the blank group (P<0.05).4. After14days of treatment, compared with model group, moxibustion group rat rectal temperature increased significantly (P<0.05), was still lower than that of the blank group (P<0.05).5. Compared with the blank group,model group, kidney weight and coefficient were significantly decreased (P<0.01), kidney weight of the moxibustion group was lower than that of the model group significantly increased (P<0.01), kidney coefficient than those in the model group significantly increased (P<0.05), but was still significantly lower than the black group (P<0.01).6. Model group rats adrenal weight and coefficient compared with the blank group were significantly decreased (P<0.01), and compared with model group, the moxibustion group were significantly increased (P<0.01), but still significantly lower than the black group (P<0.01).7. The contents of CRH in hypothalamus of rats in the model group was significantly lower than that of the black group and control model group (P<0.01), moxibustion group was increased than model group (P<0.05), but significantly lower than that of tblack group (P<0.05).Conclusion:Moxibustion on Guanyuan, Shenshu can increase the body weight of rats in growth rat, rase rectal temperature, epromote the hypothalamus of rats with deficiency of Kidney Yang release CRH, relieve the kidney, adrenal structure damage, increase the weight of adrenal gland and adrenal coefficient, which may improve the symptoms of kidney yang deficiency was its mechanism, and improve the hypothalamic pituitary gland axis on the excitability of the kidney were closely related, it was worth further deep research. |
