Effect Of Yiqi Yangyin Drugs On The Cell Model Of Deficiency Of Qi And Yin In Diabetes Mellitus

Objective:Explore the establishment of a syndrome cell model of qi and yin deficiency syndrome induced by diabetes in vitro,and observe the effect of Yiqi Yangyin medicine and ethanol extract of Coreopsis tinctoria on syndrome cell model.Method:1.Induced by the serum of patients with diabetic qi and yin deficiency syndrome and the drugs of breaking qi and damaging yin,a syndrome cell model was established and the morphology and activity of Hep G2 cells were observed.2.Using ~1H-NMR technology to detect metabolites in cells and culture supernatants of each group,and analyze pathways and enrichment.3.FOXC2m RNA expression was detected by RT-PCR,Akt/m TORproteinexpressionwasdetectedby Western-bloting.Results:1.The cell models were induced with serum(0.5%)and the decoction of qingpifuzi(0.35mg/m L),the serum and drugs had a certain inhibitory effect on cell activity and shrink in shape after 24h intervention of drug.Intervention with Yiqi Yangyin medicine and ethanol extract of Coreopsis tinctoria improves the activity of syndrome cells and makes the cell morphology close to normal.2.The experimental sample points all fall within the 95%confidence interval,and the clustering distribution between the normal and model groups is significant and the credibility is high;The OPLS-DA diagram shows that the Huangqi(Astragalus)and Shengmai groups are gradually away from the model group and gradually approach the normal group,and the ethanol extract of Coreopsis tinctoria group is away from the model group and away from the normal group;Metabolite analysis revealed differences in lactic acid,fatty acid,α-glucose and isoleucine(VIP>1 and P<0.05);Metabolite pathway and enrichment analysis results showed that were related to amino acid metabolism pathway and energy metabolism pathway.3.FOXC2m RNA transcription and p-m TOR and p-Akt protein expression levels were up-regulated in the Qi-Yin deficiency syndrome cell model group.Gene expression and protein expression in the syndrome model group were down-regulated after Shengmai intervention(P<0.05).Conclusion:The Yiqi Yangyin medicine improves the morphology and activity of the syndrome model cells,and has a certain effect on the metabolite of the syndrome model of diabetic qi and yin deficiency syndrome.
