Objective:The rhubarb liquorice decoction which has a wide range of clinical application is a classical compound from the Synopsis of Golden Chamber,the efficacy of purge and anti-nausea of it is clear through the generations.The studies in the active components alignment of the rhubarb liquorice decoction are less,so we can’t control the quality of it from the aspect of effective substances.Under the guidance of the traditional Chinese medicine compatibility theory and the active components alignment theory,the objective of this research is studying the active components alignment of the classical compound rhubarb liquorice decoction,pinpointing its corresponding relation between the ACA and compatibility.This research can provide experimental support for exploring the relationship between the "compatibility-the ACA" of traditional Chinese medicine compound,provide a scientific basis for establishing the quality evaluation system of the corresponding ACA of the Rhubarb Licorice Decoction,and lay the foundation of the development of science of TCM active components alignment and component identification.Methods and contents:In order to study the ACA of the rhubarb liquorice decoction,we choose the rhubarb liquorice decoction as the research object.By means of literature searches,sort the chemical composition and pharmacological activity with corresponding functions and indications of the RLD and its compatibility pieces Radix et Rhizoma Rhei Palmati and Radix et Rhizoma Glycyrrhizae Uralensis.Screen active ingredients as reference materials,the active ingredients must conform to the characteristics of the dosage form,have clear pharmacological actions,and correspond to the purgative and antiemetic efficacy.Compare the differences between the rhubarb and liquorice prescriptions with the different single herbs,compatibility pieces or proportion,and prepare test samples according to the traditional method.Use HPLC-DAD wavelength multiplexing method to determinate the contents of twelve different active components:sennoside B-sennoside A-aloe emodin-rhein-emodinchrysophanol-physcion,liquiritin-isoliquiritin-liquiritigenin-isoliquiritigenin-glycyrrhizic acid.Analyze the changes in the ACA of rhubarb liquorice decoction contents and proportion when the compatibility is changed,further analyze the relationship between compatibility and the AC A.The research contents include:1.the determination and analysis of ten different batches Radix et Rhizoma Rhei Palmati and Radix et Rhizoma Glycyrrhizae Uralensis ACA,2.the correlation analysis of the AC A of rhubarb liquorice decoction and its compatibility pieces,3.the analysis of the influence of the compatibility pieces on the ACA of rhubarb liquorice decoction,4.the analysis of the influence of single factor Radix et Rhizoma Rhei Palmati dosage change on the rhubarb liquorice decoction ACA,5.the analysis of the influence of Radix et Rhizoma Glycyrrhizae Uralensis dosage change on the rhubarb liquorice decoction ACA.Results:1.The content and proportion of ACA of ten different batches of Radix et Rhizoma Rhei Palmati and Radix et Rhizoma Glycyrrhizae Uralensis goods have obvious differences,the results were statistically significant(P<0.05).The content of seven ACA in ten batches born rhubarb was(μg/g):1302.46±2.71~3479.01±8.40,proportion trends of seven ACA were similar,rhein content was the highest,content of physcion was the lowest one.The content of five ACA in ten batches born licorice was(μg/g):10038.72±44.91~19656.93±12.22,proportion trends of five ACA were also similar,glycyrrhizic acid content was the highest,followed by liquiritin,isoliquiritin,liquiritigenin,isoliquiritigenin.2.After the compatibility of Radix et Rhizoma Rhei Palmati and Radix et Rhizoma Glycyrrhizae Uralensis,the content and proportion of their ACA changed significantly(P<0.05),content of bianthrone component decreased significantly,free anthraquinones component content less obvious changed,the contents of glycyrrhizic acid and licorice flavonoids were reduced to varying degrees.The ACA corresponding to the purgative and antiemetic efficacy of the RLD which comprised Radix et Rhizoma Rhei Palmati produced in Shanxi(batch 150204)and Radix et Rhizoma Glycyrrhizae Uralensis produced in Inner Mongolia(batch 150204)were:(μg/mL,with the test solution the content meter):sennoside B(2.490±0.012)-liquiritin(25.161± 0.710)-sennoside A(3.453 ± 0.141)-isoliquiritin(2.305±0.158)-liquiritigenin(4.922±0.080)isoliquiritigenin(0.308 ± 0.004)-aloe emodin(4.281 ± 0.122)-glycyrrhizic acid(36.469± 0.465)-rhein(22.212 ± 0.487)-emodin(3.877±0.019)-chrysophanol(2.664 ± 0.066)-physcion(1.025±0.020).3.Compatibility ratio is fixed at Radix et Rhizoma Rhei Palmati:Radix et Rhizoma Glycyrrhizae Uralensis 12g:3g,change the compatibility pieces,the ACA of Radix et Rhizoma Rhei Palmati,Radix et Rhizoma Glycyrrhizae Uralensis and the total content of 12 kinds of ACA in the RLD were changed significantly(P<0.05),wherein the variation of the total amount of ACA were:Born rhubarb:born licorice>born rhubarb:Zhigancao>cooked rhubarb:born licorice>rhubarb stir-fried with wine:born licorice>cooked rhubarb:Zhigancao>rhubarb charcoal:born licorice>rhubarb stir-fried with wine:Zhigancao>rhubarb charcoal:Zhigancao.The degree of influence of each compatibility pieces on the ACA:born rhubarb>cooked rhubarb>rhubarb stir-fried with wine>rhubarb charcoal;born licorice>Zhigancao.4.Compatible dose of single factor Radix et Rhizoma Glycyrrhizae Uralensis changed,the ACA of Radix et Rhizoma Rhei Palmati,Radix et Rhizoma Glycyrrhizae Uralensis and the total content were changed significantly(P<0.05).Compatible dose of Radix et Rhizoma Rhei Palmati stays 12g unchanged,when doses of Rhizoma Glycyrrhizae Uralensis were 0.1g,0.3g,1g,3g,6g,9g,12g,the total content of ACA of the RLD were(μg/g):2259.49±7.49,2870.58±0.14,4034.58±7.74,5475.56±32.00,8541.69±3.42,10370.45±6.72,10707.21±13.98.When the dose of Rhizoma Glycyrrhizae Uralensis was increased,the content of Radix et Rhizoma Rhei Palmati ACA decreased,the total amount of ACA of the RLD increased.5.Compatible dose of single factor Radix et Rhizoma Rhei Palmati changed,the ACA of Radix et Rhizoma Rhei Palmati,Radix et Rhizoma Glycyrrhizae Uralensis and the total content were changed significantly(P<0.05).Compatible dose of Radix et Rhizoma Glycyrrhizae Uralensis stays 3g unchanged,when doses of Radix et Rhizoma Rhei Palmati were 3g,6g,9g,12g,15g,18g,21g,the total content of ACA of the RLD were(μg/g):10871.94±0.19,8831.31± 1.57,7205.46±8.77,5475.56±32.00,5296.45±21.05,5118.08±7.11,4606.44± 11.42.When the dose of Radix et Rhizoma Rhei Palmati was increased,the content of Radix et Rhizoma Glycyrrhizae Uralensis ACA decreased,the total amount of ACA of the RLD decreased.Conclusion and discussion:1.This research established the HPLC-DAD multi-wavelength components determination method of 12 kinds of the ACA corresponding to the purgative and antiemetic efficacy of the RLD and its compatibility pieces(sennoside B-sennoside A-aloe emodin-rheinemodin-chrysophanol-physcion,liquiritin-isoliquiritin-liquiritigenin-isoliquiritigeninglycyrrhizic acid).The method is easy to operate,has good reproducibility and specificity,and has scientific and feasible when used for the quality control of the RLD.2.Content and proportions of the ACA of different batches of Radix et Rhizoma Rhei Palmati and Radix et Rhizoma Glycyrrhizae Uralensis have a significant difference.This suggests us that using the ACA as reference materials for measuring the quality of rhubarb,licorice or the RLD and establishing the quality standard of the RLD needs to determine and analyze a large number of sample batches of Radix et Rhizoma Rhei Palmati,Radix et Rhizoma Glycyrrhizae Uralensis,then sum up the content and proportion range of the RLD.To investigate the relationship between the ACA of RLD and compatibility,we should choose the same batch of Radix et Rhizoma Rhei Palmati and Radix et Rhizoma Glycyrrhizae Uralensis to eliminate interference between batches produced.Compatibility has significant impact on the content,proportion and the total content of ACA of the RLD.The compatibility pieces play a part in the ACA,after the compatibility of Radix et Rhizoma Rhei Palmati and Radix et Rhizoma Glycyrrhizae Uralensis,the content and proportion of their ACA changed significantly,this proved that ACA of Chinese medicine compound are not a superposition of ACA of its compatibility pieces,there exists physical or chemical interaction between its compatibility constituent,and the interaction changed the dissolution of ACA.When the compatibility proportions were the same and the compatibility pieces were different,the content of ACA was different.When the compatibility pieces were born rhubarb and born licorice,content of ACA was the highest,proved the science of classical prescription.When compatibility pieces were the same,changed the compatibility doses and proportions,the content of ACA were also different,there is one-to-one relationship between ACA and compatibility proportion.And the influence of Radix et Rhizoma Glycyrrhizae Uralensis on AC A of the RLD is stronger than Radix et Rhizoma Rhei Palmati,explained that licorice has a very important impact on the efficacy of RLD,its role cannot be ignored although licorice is the minister drug,in addition this conclusion is also possibly connected with the ACA we screened were not fully.The clinical efficacy will change when ACA is not the same,suggesting that we should pay attention to varieties,processed products and doses of compatibility pieces,and be cautious when prescription and dispensing.When compatibility pieces and proportion are certain,the ACA of Chinese medicine compound is also certain,proving that using the ACA with specific content and proportion of the RLD as reference materials for its evaluation and quality control on clinic is scientific.
