Objective:Blumea balsamifera DC.is a perennial herb.Its whole plant contains flavonoids,volatile oil and other compounds,and has anti-tumor,anti-oxidant and other pharmacological activities.The main active fraction were summarized in the literature.The dichloro methane extacts,this experiment mainly uses a variety of chromatographic methods to separate and purify the Blumea balsamifera dichloro methane extacts and then investigate the anti-non-small cell lung cancer cell activity of the isolated compound.Methods:1?Using ethanol reflux to extract the dried whole plant of Blumea balsamifera to obtain extract,Separation and purification the dichloro methane part of Blumea balsamifera by liquid-liquid extraction,silica gel column chromatography,C 18 reverse phase column chromatography and semi-preparative chromatography.The structure of the isolated pure compound was identified by UV,ESI-MS,NMR;finally,the structure was analyzed by MestReNova software to obtain the pure compound.2?The CCK-8 method was used to detect the inhibitory activity of A549 and NCI-H1299 on the non-small cell lung cancer cell line A549 and NCI-H1299 by using the CCK-8 method to detect the extraction site and compounds.The IC50 value of the half inhibition rate was calculated by SPSS,and the positive drugs were analyzed and compared to inhibit A549 and NCI-H1299 results.Result:1?A total of 13 pure compounds were separated,and their struct ures were analyzed according to NMR.Compound ?(5,4'-dihydrox y-7,3-dimethoxyflavonoid),compound ?(5,7-dihydroxy-3,3',4'-Trimet hoxyflavonoid),compound ?(5,7-dihydroxy-3,4'-dimethoxyflavonoi d),compound ?(Ursolic acid),compound V(Oleanolic acid),com pound ?(5-hydroxy-3,7,4'-Trimethoxyflavonoid),compound ?(Pr otocatechuic acid),compound ?(Caffeic acid).Among them,co mpound ????????? are the first isolation from this plan t.2?The dichloro methane part of Blumea balsamifera has a significant inhibitory effect on A549 and NCI-H1299 in non-small cell lung cancer cells,and is concentration-dependent.The IC50 values for A549 and NCI-H1299 are:545.659?g/ml and 243.942?g/ml.3?Compared with other compounds,compound ?,compound ?and compound ? have significant inhibitory effects on two types of non-small cell lung cancer cells.The IC50 values of these three compounds on A549 cells are 6.885 ?M,204.431 ?M and 75.245 ?M,respectively.,The IC50 values of the three compounds on NCI-H1299 cells are 61.287?M,69.013 ?M and 28.149?M respectively.Compounds ?,?,? and ? also have a certain concentration-dependent inhibitory trend on the two cells.Conclusion:A total of 13 pure compounds were separated,and their structure s were analyzed according to NMR.Compound ?(5,4'-dihydroxy-7,3-dimethoxyflavonoids),and compound ?(5,7-dihydroxy-3,3',4'-Trim ethoxyflavonoids),compound ?(5,7-dihydroxy-3,4'-dimethoxyflavono ids),compound ?(ursolic acid),compound ?(oleanolic acid),co mpound ?(5-hydroxy-3,7,4'-Trimethoxyflavonoids),compound ?(protocatechuic acid),compound ?(caffeic acid).Among them,compound ????????? is the first isolation from this pla nt.The dichloro methane part of Blumea balsamifera has a certain inhibitory effect on two kinds of non-small cell lung cancer cells,and in the in vitro activity detection of monomer compounds,it is concluded that compound ? and compound ? are compared with other compounds on two non-small cell lung cancer cells.Small cell lung cancer cells have obvious inhibitory activity,which provides a certain preliminary experimental basis for subsequent in vivo experimental studies.
