| Objective: Selaginella tamariscina,a dried whole grass of the genus of Selaginella,is a national standard medicinal material indexed in the Chinese Pharmacopoeia.Due to its unique biological characteristics and the structural specificity and diversity of its active constituents,selaginella has attracted many domestic and foreign researchers attention.This dissertation mainly conducts systematic research on the chemical constituents of Selaginella tamariscina,and discovery secondary metabolites with novel structures,especially novel alkyne phenols and their derivatives,and evaluates bioactive compounds with anti-hepatic cancer activity through in vitro screening.Methods: The 75% ethanol extract of selaginella tamariscina was separated and purified by modern chromatographic separation technology including silica gel column chromatography,dextran gel chromatography,ODS reversed-phase column chromatography,and preparative high-performance liquid chromatography.The structrue of these compounds were confirmed by 1D / 2D NMR and high resolution mass spectrometry.CCK8 experiment was used to screen the obtained compounds for anti-liver cancer activity in vitro,and the effects of monomer compounds on the viability of human liver cancer SMMC-7721 cells and human liver cancer Hep-G2 cells were evaluated,and the anti-liver cancer effects of several compound were found.Results: Twenty-four compounds were isolated included thirteen alkyne phenols and their derivatives,one novel skeleton type compound,one diphenylindenone,one benzophenone,one benzofuranone,one acetylenone compound.,one lignan,two flavonoids,and three simple phenolic acid compounds.compounds 1*-8* are new compounds,in which compound 1* is a new compound with a phenylfluorene-naphthalenespirocyclohexadienone skeleton-type and compound 2* is an alkyne phenols derivative with a novel skeleton-type,compound 3*-5* is a new alkyne phenols derivatives,compound 6* is a diphenylindenone rare in nature,compound 7* is the first diphenylbenzophenones found in nature,compound 8* is biphenyl benzofuranones.In addition,compound 18 and compound 19 were isolated from Selaginella tamariscina for the first time.In vitro human anti-liver cancer screening revealed that compound 11 and compound 18 have significant proliferation inhibitory activity on two liver cancer cell lines,and compound 18 has better cytotoxic activity.Conclusions: A total of twenty-four compounds were isolated and identified including 8 new compounds.Five compounds with anti-hepatocellular carcinoma activity were screened out,and compounds11 and 18 had the best inhibition.. |
