| Objective:Depression and hypertension interact with each other,forming a vicious circle that worsens the condition.The purpose of this study is to explore the medication use of TCM in the treatment of Hypertension with depression,to predict the potential mechanism of action of core drug combinations,to provide data basis and reference for subsequent research and clinical applications.Methods:Literature on the treatment of hypertension combined with depression by traditional Chinese medicine in Chinese and English database was searched and summarized to construct the prescription database of hypertension combined with depression.The traditional Chinese medicine included in the database was analyzed by drug classification and the frequency of property,taste and meridian tropism.SPSS Clementine 12.0 software was used to analyze the association rules of drugs,and principal component analysis and inter-group linkage method were used for factor analysis and systematic cluster analysis.Using "depression" and "hypertension" as keywords,the targets related to drugs and hypertension combined with depression were obtained from TCMSP database and OMIM database,respectively.The intersection targets were uploaded to the STRING database to construct a protein-protein interaction network.Cytoscape 3.7.2 software was used to calculate the Network topology parameters and obtain the key targets by using the Network Analyzer tool.Finally,the key components and targets were reverse verified by molecular docking technology.Results:Through bibliometrics and data mining,this study found that the elderly had a high incidence of hypertension combined with depression,and Guipi decoction was the most concerned TCM compound.Network pharmacology was used to predict the potential targets and pathways of core drug combinations.A total of 1568 disease targets of hypertension and 2124 disease targets of depression were obtained,and 248 common gene targets were screened.The core targets included INS,IL6,STAT3,TNF,AKT1,VEGFA,ALB,MAPK3,EGFR,and JUN.The core medicinal chemical components included quercetin,stigmasterol,kaempferol,adenosine triphosphate,and zingerone.GO functional enrichment analysis showed that biological processes mainly included positive regulation of gene expression and response to lipopolysaccharide,while KEGG enrichment pathways mainly included neuroactive ligand-receptor interaction signaling pathway,MAPK signaling pathway and calcium signaling pathway.Conclusion:The core drugs of traditional Chinese medicine in the treatment of hypertension combined with depression are mainly from Guipi decoction.To determine the mechanism of interaction between Guipi decoction and Chinese herbs is helpful to better treat and prevent hypertension combined with depression and improve the health of patients. |
