Objective:Carry out data mining on the medical records of treating acute heart failure with traditional Chinese medicine,explore its medication rules,refine the core formula,conduct network pharmacology research on it,and further clarify the mechanism of the core formula in treating diseases.It is expected to provide basis and new research ideas for the treatment of this disease,provide new support for clinical rational drug use,and provide reference for later clinical treatment and new drug development.Methods:This paper mainly searches the clinical randomized controlled trials of all the Chinese herbal compounds in the existing database from January 2002 to January 2022 in the treatment of acute heart failure,collects the Chinese herbal compounds with definite therapeutic effects,and imports the data into the cloud platform of ancient and modern medical records after standardized processing,and carries out the drug frequency of the included Chinese herbal drugs through statistical analysis,cluster analysis,association analysis,complex network analysis and other methods in the "data mining"module The efficacy,sex and taste,meridian tropism,common compatibility and core formula are analyzed.Search the effective active ingredients and corresponding protein targets of eight traditional Chinese medicines in the core formula through the TCM system pharmacological database and analysis platform(TCMSP),and standardize the gene names of all the targets through the UniProt database.Search disease genes in Gene Cards database and OMIM database.The disease-related gene target and drug-related gene target are combined and then intersected.The intersected target is imported into SRING data to build a protein mutual aid network(PPI),and the core target gene of the drug acting on the disease is obtained.The drug corresponding target and disease corresponding target are imported into Cytascape to visualize the interaction of drug active component-disease-target.Import the intersection target of drugs and diseases into DAVID database(conduct GO and KEGG enrichment analysis to obtain the main action pathway of target gene target.Results:①A total of 109 documents meeting the requirements were retrieved,including 109 traditional Chinese medicine prescriptions and 130 traditional Chinese medicine.According to the analysis of the drug use frequency,the top 10 drugs with the most use times are Astragalus,Poria cocos,Cinnamomum cassia,Salvia miltiorrhiza,Atractylodes macrocephala,Ligusticum chuanxiong,Semen dracae,roasted licorice,aconite and ginseng.The main meridian of drugs is lung meridian,followed by heart meridian,spleen meridian,liver meridian and kidney meridian.The main flavor of the medicine is sweet,followed by pungent,bitter and mild.The use of moderate temperature is the highest,followed by mild,mild and mild temperature.The drug pairs that highly support the association rule include Poria coco-Cinnamomum twig,Astragalus membranaceus-Salvia miltiorrhiza,Atractylodes macrocephala-Poria cocos,Chuanxiong-Salvia miltiorrhiza,and Tinglizi-Poria cocos.After data mining,the core formula contains 8 drugs,including cinnamon twig,poria cocos,astragalus,salvia miltiorrhiza,atractylodes macrocephala,ligusticum chuanxiong,aconite,and rhizoma scapulosa.High-frequency traditional Chinese medicine can be divided into four categories.The first category is Semen Lepidae and Radix Glycyrrhizae;The second category:cassia twig,tuckahoe,atractylodes macrocephala;The third category:aconite and ginseng;Class IV:Chuanxiong,Huangqi,Danshen.② 145 active ingredients of core drugs have been screened,and the main active ingredients that play a key role are quercetin,luteolin,kaempferolβ-Sitosterol,isorhamnetin,tanshinone IIA,etc.The core targets to play a role are mainly AKT,STAT3,MAPK1,MAPK14,TNF,IL-6,MAPK8,etc.The signal pathways involved in regulation include PI3K-Akt signal pathway,mTOR signal pathway,mitochondrial autophagy,PPAR signal pathway,TNF signal pathway,HIF-1 signal pathway,etc.Conclusion(s):1.In the treatment of acute heart failure,the prescription should be based on the mild and sweet taste,and the mild medicine should be used.The bitter and bitter taste should be dry and damp,and the medicine with severe cold and heat should not be used.The main meridian is lung meridian,followed by heart meridian and spleen meridian.2.According to data mining,the core prescription of TCM compound prescription for treating acute heart failure is cassia twig,poria cocos,astragalus,salvia miltiorrhiza,atractylodes macrocephala,ligusticum chuanxiong,aconite,and scallion.3.The core formula contains a variety of effective active ingredients,which can regulate the disease at multiple targets and participate in multiple signal pathways to play the role of treating acute heart failure.4.The mechanism of the core prescription in treating acute heart failure may be related to inhibiting cell apoptosis,reducing myocardial fibrosis,reducing oxidative stress response,promoting cell oxidative energy supply,reducing myocardial cell damage,enhancing myocardial contractility,improving myocardial metabolism,etc.
