| Purpose:This study uses metabonomics technology to analyze the serum samples of patients with Qi and yin deficiency syndrome of metabolic syndrome and healthy people,try to find the characteristic biomarkers of Qi and yin deficiency syndrome of metabolic syndrome,further explore the molecular mechanism of Qi and yin deficiency syndrome of metabolic syndrome from the perspective of metabolites,and reveal the biological essence of TCM syndrome of Qi and yin deficiency syndrome of metabolic syndrome.Material and method:Through strict screening,36 patients with deficiency of Qi and Yin of metabolic syndrome(group B)were hospitalized in the Department of endocrinology of the Affiliated Hospital of Liaoning University of traditional Chinese medicine,and 32 healthy people(group A)were in the control group.All personnel signed the informed consent form.This research scheme was approved by the ethics committee of the Affiliated Hospital of Liaoning University of traditional Chinese medicine.The collected serum samples were separated by ultra high performance liquid chromatography(UHPLC),and then analyzed by mass spectrometry.Convert the original data,and use msdial software to align the peaks,correct the retention time and extract the peak area.The metabolites were identified by accurate mass number matching(< 25ppm)and secondary spectrum matching,and the databases such as massbank and HMDB were searched.Delete the ion peak with the missing value > 50% in the obtained data group,integrate the positive and negative ion peaks,and apply the software simca-p14 1 for pattern recognition,use unit variance scaling(UV)to preprocess the data and conduct multidimensional statistical analysis.Results:1.In OPLS-DA pattern discrimination diagram,the samples of Qi Yin deficiency syndrome group and healthy control group showed aggregation distribution within the group respectively;The samples of Qi Yin deficiency syndrome group and healthy control group are respectively distributed on both sides of the central axis.The samples of the two groups are clearly distinguished in the figure,indicating that there is a significant difference in metabolic status between patients with Qi Yin deficiency syndrome of metabolic syndrome and healthy people.2.The comparative study of two groups of serum samples found that 11 different metabolites were identified,namely pyruvate,N-acetylneuraminic acid,L-lysine,L-glutamine,taurine,valine,phenylalanine,L-tyrosine,choline,arachidonic acid and hypoxanthine.Differential metabolic pathways involve amino sugar and nucleotide sugar metabolism;Protein digestion and absorption;Taurine and sub taurine metabolism;Amino acid metabolism;Valine,leucine and isoleucine metabolism;Phenylalanine metabolism;Glycerol phospholipid metabolism;Biosynthesis of unsaturated fatty acids;Purine metabolism;Alanine,aspartic acid and glutamate metabolism.Conclusion:1.Using metabolomics technology to analyze the serum samples of patients with Qi Yin deficiency syndrome of metabolic syndrome,the metabolite map obtained is obviously different from that of healthy people,and the potential biomarkers related to patients with Qi Yin deficiency syndrome of metabolic syndrome are found.2.Find the potential biomarkers of patients with Qi Yin deficiency syndrome of metabolic syndrome such as pyruvate,N-acetylneuraminic acid,L-lysine,L-glutamine,taurine,valine,phenylalanine,L-tyrosine,choline,arachidonic acid and hypoxanthine,and reveal that the occurrence of Qi Yin deficiency syndrome of metabolic syndrome is related to metabolic disorders such as sugars,proteins,lipids and purines,These small molecular compounds may be potential targets of TCM Syndromes of Qi Yin deficiency syndrome of metabolic syndrome.3.Metabonomics technology can explore and study the scientific essence of traditional Chinese medicine from the perspective of metabolites.It is one of the effective methods to reveal the essence of TCM Syndromes of diseases. |
