Purpose:To explore the effect of electroacupuncture at Shenmen,Sanyinjiao,Ganshu and Taichong points on the serum of insomnia rats with liver depression IL-1β、NEE and the influence of MT content in hypothalamus,and the influence on animal behavior of rats;It also preliminarily explains the mechanism of electroacupuncture "Ganshu" on the recovery of sleep-wake cycle disorder in rats with insomnia due to liver depression.Material and method:1.12 male SD rats were randomly selected as blank group,The remaining 36 rats were subjected to tail clipping stimulation combined with intraperitoneal injection of PCPA model replication.After 16 days of modeling,the rats were randomly divided into model group,acupuncture group,and western medicine(diazepam)group.In the blank group,weak alkaline saline was injected on the 15 th day for 2 days.The normal group and the model group were given no treatment except grasping and fixing at the same time.The electroacupuncture group was treated with electroacupuncture at Shenmen,Sanyinjiao,Ganshu and Taichong points.The western medicine group was treated with diazepam tablet solution by gavage for 6 days.After the treatment,the rats’ mental state and weight changes were observed.The elevated cross maze test and open field test were used to detect and record the OE and OT values,horizontal scores,vertical scores,and fecal particles of rats in each group.2.Detection of rat serum IL-1 by enzyme-linked immunosorbent assay(ELISA)β、NE and MT content in hypothalamus,and analyze data。Results:1.Behavioral observation: Before the model was established,the rats worked and rested regularly,sleeping in the daytime and rising at night.The hair is glossy and the two stools are normal;After modeling,the rats were depressed,their hair was dry,dull,their diet was reduced,their vigilance increased,and their aggressiveness increased.Compared with the blank group,the body weight of rats in the model group。After treatment,compared with the blank group,the weight of the model group,the electroacupuncture group and the western medicine group increased,with a statistically significant difference(P < 0.05)。The results of the elevated cross maze test show that,OE% Compared with the blank group,the value of the model group is significantly reduced(P < 0.05),Compared with the model group,the number of electroacupuncture group and western medicine group increased significantly(P< 0.05)。OT% Compared with the blank group,the model group,electroacupuncture group and western medicine group all decreased(P < 0.05),Compared with the model group,the electroacupuncture group and western medicine group increased(P < 0.05)。The results of the open field experiment showed that compared with the normal group,the horizontal score was significantly reduced in the model group(P < 0.05),After treatment,compared with the model group,the electroacupuncture group and the western medicine group were increased(P < 0.05)。Vertical score comparison,compared with blank group,the model group is significantly different(P < 0.05);Compared with the model group,there were obvious differences between the electroacupuncture group and the western medicine group(P< 0.05)。Compared with the blank group,the number of fecal particles was different from that of the model group and the western medicine group(P < 0.05);Compared with the model group,the difference between the electroacupuncture group and the western medicine group was statistically significant(P < 0.05)。2.After electroacupuncture,serum IL-1β,NE and MT expression in rats was shown(1)IL-1βnet contents Compared with the blank group,the IL-1β content of rats in the model group was significantly increased(P < 0.05);Compared with the model group,the IL-1β content of the electroacupuncture group and the western medicine group decreased significantly(P <0.05)。(2)NE net content Compared with the blank group,the NE content of rats in the model group was significantly increased(P < 0.05);Compared with the model group,the NE content in the electroacupuncture group and the western medicine group decreased significantly(P <0.05);Compared with the Western medicine group。(3)MT net content Compared with the blank group,the IL-1β content of rats in the model group was significantly reduced(P < 0.05);Compared with the model group,the electroacupuncture group was compared with the western medicine group IL-1β significantly increased(P <0.05);Compared with the western medicine group,the electroacupuncture group IL-1βHigher levels of content(P < 0.05)。Conclusion:1.Electroacupuncture at Shenmen,Sanyinjiao,Ganshu and Taichong points can improve the sleeping state of insomnia rats and reduce anxiety effectively.2.Electroacupuncture at Shenmen,Sanyinjiao,Ganshu and Taichong points can reduce IL-1βand NE content and increase MT content in hypothalamus of insomnia rats.At the same time,electroacupuncture Shenmen,Sanyinjiao,Ganshu,Taichong point to ease the liver Qi,balance Yin and Yang,providing a theoretical basis for the intervention of liver depression insomnia.
