There are three chapters in this thesis. In Chapter I, the principle of molecularimprinted polymers(MIPs), polymerization methods, application of this technique and itsfuture developments were introduced. Meanwhile, the applications of HPLC on theanalysis of anti-cancer drugs were also described.Chapter II: Molecular imprinted polymers. First, molecular imprinted polymers (MIPs) by using single template(pentoxifhylline, caffeine and theophylline) and mixed-templates (pentoxifhylline-theophylline and caffeine-theophylline) were investigated in this chapter; Second, Caffeine molecular imprinted uniform microgel spheres were prepared by precipitation polymerization method, comparison of caffeine adsorption on molecular imprinted and blank microgel spheres was made and Scatchard analysis was used to investigate the binding properties of caffeine on imprinted microgel spheres. Third, the molecular imprinted technique was applied for the preparation aminoantipyrine imprinted polymer stationary phase. The adsorption isotherms of aminoantipyrine, antipyrine, pyrazolone and the competitive adsorption isotherms of aminoantipyrine and antipyrine on the imprinted stationary phase were determined using rectangular pulse frontal analysis and static method.Chapter III: Separation and analysis of natural and genetic anti-cancer drugs by HPLC. In this chapter, supercritical fluid extraction (SFE) with co-solvent scheme for the extraction of Acanthoside-D from Acanthopanax senticosus was developed. Different pressure and water content in the fluid were investigated to see their effects on extraction results; Second, the optimum operating conditions were experimentally determined in the analytical and preparative recycle chromatography for the extraction and preparation ofacanthoside-D from Acanthopanax Senticosus. Apart from the above study, high-performance frontal analysis (HPFA) was used for protein binding study of perillyl alcohol (POH) enantiomers to human serum albumin (HSA) , a RP-HPLC method was developed for the separation and purification of recombinant human interleukin-18(IL-18) on C8 column, and a CE method for the separation and determination of the active anthraquinone components, physcion, chrysophanol, aloe-emodin, emodin, and rhein in Chinese herb Polygnum multiflorum Thunb. were developed.
