With the fast development of biotechnology, the life science has got substantial achievement. The complex interactions between biomolecules in organisms compose the fundamental units for life processes. The exploration of the life essence means the characterization of corresponding biomolecular interactions. The investigation of the! molecular interactions is the key groundwork in modern high throughput drug screening (HTS). And that, capillary electrophoresis (CE) is an important technique in the study of biomolecular interactions. Since the past ten years, analyses of many binding systems have been accomplished by CE. The methodology of CE has obtained notable progresses.Based on the interaction systems mainly containing DNA, this dissertation used CE to characterize both DNA-macromolecule and DNA-small molecule bindings. Particularly, CE was served to probe the chiral recognition of DNA to drug enantiomers. The present paper was the first report concerning the investigation of binding discrimination of DNA to the enantiomers of non-metal organic complex by CE. Capillary zone electrophoresis (CZE) is the most sample-saving one among the five CE modes that have been used to study molecular interactions. In case of only very small amount of samples available, e.g. in some HTS which need scarce targeting molecules, CZE is trie most appropriate separation mode. CZE can be successfully applied to weak binding systems in the comparison of different operation modes, whereas, it was considered that CZE was not suitable to study this type of binding system before. The feasibility of CZE in evaluating weak binding systems through the studies of both macromolecule-macromolecule and macromolecule-small molecule interactions were shown in our work. This could broaden the applications of CZE in the area of large scale HTS.
