Based on a large number of resources investigation and literature systematization, the medical bioactive ingredients of Marchantia convoluta living in Guangxi China were made as object. During pharmacological research, the chemical compositions of the plant were analyzed, extracted, separated and purified, and the crude drug identification was engaged. Then some effective bioactive ingredients - flavones were obtained which possessed the action of anti-hepatitis B virus, protection of liver function, antibiosis, anti-inflammation and diuresis. The qualitative and quantitative analysis were carried out by means of HPLC, UV, IR, HNMR, MS, and HPLC-MS to determine the structure of the monomers and set up the theoretical separating model of the flavones solid-phase extraction. Moreover, the monomers were prepared into injection. Details are as following:1. Investigation and identification of the plant samples. Three species of Marchantia plants in Guangxi (Marchantia polymorpha, Marchantia convoluta and Marchantia paleacea) were identified through investigation of producing area, collection of samples and identification of crude drug. These plants were distinguished through comparing their shape and characteristics, microscopic structure, ultraviolet spectra and fingerprint chart. The type of chemical composition in Marchantia convoluta were primarily found out, the essential oil was analyzed by GC-MS, and two sterol compounds (B-sistosterin and stigmasterol) were yielded by separating the lip soluble part with silica-gel column chromatography.2. Assaying of total flavones. The contents of total flavones in Marchantia convoluta were determined through developed visible spectrophotometry. By studying the factors that affected the determination, the optimum conditions for this experiment were found: NaNO2-AlCl3, color-developing agent; 15 minutes, color time; 525nm, wavelength. The data of the content and absorbance formed a standard curve, namely Y= -0.0153+0.03003X; the recovery of the samples was 97.61% to 101.59%; the average recovery was 99.28%, and the precision was high (RSD=1.1%, n=5). The operation was easy, fast and reliable, and it can be used to determine total flavones.3. Extraction of total flavones. After carrying out an orthogonal test to study the affected factors of extraction rate, the best extraction conditions forIVtotal flavones in Marchantia convoluta was discovered as follows: temperatures, 70 癈; time, 30 min; times, 3; solvent, 80% alcohol; material and ratio of solvent, 1:15; and the yield rate, 1.90%. Before and after being dealt with enzyme , the extract rate was improved with 4.59%.4. Separation and identification of flavones monomer. Flavones monomer was prepared through solid-phase extraction, and the purity determination was on process with HPLC at the same time. After determining the structure by means of physic-chemical method combined with the application of UV, IR, HNMR, MS and HPLC-MS, three monomers were obtained: 5-oxhydryl-7-methoxyl-2-methyl chromone (I), 7-oxhydryl flavones (II), apigenin-7-O-B-D- glucuronide (III). Incidentally, the monomers I and II were separated from the species of plant for the first time.5. Analysis of flavonoid compounds with HPLC. The reversed-phase high-performance liquid chromatography was set up to determine the content of flavones compound in Marchantia convoluta. The material steps were as follows: taking acetic acid-methanol-acetonitrile- phosphoric-acid as the multicomponent mobile phase and eluting comparably, then the flavones extraction was separated and determined directly with 30 minutes. The detection wavelength was 350 nm, the flow velocity was 0.60mL/min, and the quantitative analysis was conducted with calibration curve method. The result indicated that the average recovery of samples was 96.79% to 101.13%, the relative standard deviation was 0.66% to 1.52%.
