| Fig leaves are leaves of common fig which belongs to moraceae ficus subgen ficus specy. They can be got each summer and autumn, used freshly, dried open-air or yin heavenly. The leaves are traditionly claimed to be effective in expelling dampness, anti-damp toxin, detumescence and alleviating pain. They are used in diarrhea due to damp-heat, haemorrhoids, swollen piles and scrofula. Pharmacological data validated that the extractive of fig leaves has visible activities of antioxidant, antibiosis and antiviral. As a result, it can be used to cure diabetes, antiatheroscloresis and cancer. The studies of extract from fig leaves are mainly in psoralen and bergapten. However, there has been few report on flavonoids. Therefore this study was focused on content of total flavonoids in fig leaves, abstraction, purification and physiological activity. This study obtained brown powder using ethanol as a solvent for extraction, dry fig leaves to impurities, vacuum dried, and it is identified as flavonoids after a qualitative color reaction. The content of total flavonoids is2.60%, using spectrophotometry and rutin as reference materials. The studies on optimum conditions of extraction from fig leaves, appled orthogonal experimental design method, using ethanol as extraction agent, fig leaves flavonoids in crude and yield as indexes. The optimum conditions in laboratory are70%ethanol solution as extract,1:15as solid-liquid ratio, extracting5h at70℃constant temperature water bath. And in the industrial scale, it can be70%ethanol solution as extract,1:10as solid-liquid ratio and extracting5h at70℃constant temperature water bath.Using static absorption to optimum maeroporous resin which was used for purifying total flavonoids in Fig leaves. Using dynamic absorption to the condition of purifying total flavonoids of Fig leaves by maeroporous resin. Optimum maeroporous resin was D101,and the optimum ondition was as follow:the speed that sample flows through maeroporous resin was2.0mL/in, and the total flavonoids absorbed on maeroporous resin can eluted by75%ethanol of12BV.The flavonoids extract from fig leaves were taken orally by streptozotocin diabetic mice, and its role on hypoglycemic and lipid regulating were certificated by examining the12h fasting glucose, triglycerides and total cholesterol. The antimicrobial tests in vitro of fig leaves flavonoids using filter paper method proved that Escherichia coli, Staphylococcus aureus and Bacillus subtilis are sensitive to the fig leaves flavonoids by examining the sizes of inhibition zone. The study on The Extracting of total flavones and Physiological from Fig leaves is the first time. |
