The marine invertebrates soft coral, as Colenterata Anthozoa and Alcyoniidae, were distributed over tropical or subtropical ocean zone .The South China Sea is characterized of tropical weather, contains plentiful of marine organisms.Among them,there are about 96 species soft coral. Since 1960, a lot of structurally novel and bioactive secondary metabolities, including sesquiterpenes, diterpenes, sterols, ceramides and somon, have been isolated from soft coral.The constituents exhibited potential bioactivities such as antiflammation, antitumor, antifouling and cytotoxic activities.In the continuing studies on the compounds having good bioactivities and novel structures,we investigated the soft coral Dendronephthya sp. Nephthea sp.md Sinularia sp. collected from Sanya, Hainan Province of China. The MeOH extracts of three species exhibited marktable cytotoxic activities against selected tumor lines, human leukemic cell (HL-60), human prostate cancer cell (PC-3MIE8), human gastric cancer cell (BGC-823), human breast cancer cell (MDA-MB-435), human hepatoma cancer cell (Bel-7402) and human cerrical cancer cell (Hela), respectively. Biossay-guided isolation resulted in 55 compounds, 11 new ones, as well as 44 known ones. Their structures and stereochemistry were determined on the basis of MS, IR, 'H NMR, 13C NMR and 2DNMR ('H-'HCOSY, HMQC, HMBC, 'H-'HNOESY) spectroscopic analysis and comparison with the literatures.28 Compounds, pregna-1, 4, 20-trien-3 -one (HSC-8A-P1), cholesta-4, 22(?-diene-3-one (HSC-8A-P2), 24-methylenecholesta-4, 24(28)-diene-3-one (HSC-8A-P3), 24-methyl cholesta-4, 22(?-diene-3-one(HSC-8A-P4), cholesta-4-ene-3-one(HSC-8A-P5),(24/?)-24-ethylcholesta-4-ene-3-one(HSC-8A-P6),(245)-24-ethylcholesta-4-ene-3-one(HSC-8A-P7),diiso-butyl-o-phthalate(HSC-8A-P8),2a-car bethoxy-2p-hydroxy-A-nor-cholesta-5-ene-4-one(HSC-8A-P9),24-methyl-2a-carbeth oxy-3p-hydroxy-A-nor-cholesta-5,22(E)-diene-4-one(HSC-8A-P10),3p-hydroxy-3a-c arbethoxy-A-nor-cholesta-5, 22(23, E)-diene-4-one(HSC-8A-Pll), 3p-formyloxyl-cholesta-5,22(?-diene(HSC-8B-Pl), 24-methyl-3p-formyloxylcholesta-5,22(?- di ene(HSC-8B-P2), 3p-formyloxylcholesta-5-ene (HSC-8B-P3), octadecanon-2 (HSC-8B-P4),2E-ene-octadecanal(HSC-8B-P5),cholesta-5,22-diene-3p,7a-diol(HSC -8B-P6),24-methylene-cholesta-5,22E-diene-3p,7a-diol(HSC-8B-P7),24-methylenec holesta-5, 24(28)-diene-3p, 7a-diol (HSC-8B-P8), cholesta-5-ene-3p,7a-diol(HSC-8B-P9), cholesta-5-ene-3p,7p-diol(HSC-8B-Pl0), 1,3,5-trimethyloxy-2, 4, 6-trimethyl benzene (HSC-8D-P4), A4(5)(?, A8(9)(? -sphingol-n-hexadecamide (HSC-8D-P5), cholesta-5-ene-3p-ol(HSC-8D-P6), 5a,8a-epidioxy-24-methylcholesta -6, 22(? -diene-3p-ol (HSC-8D-P7), N-hexadecanoyl-octadecasphinga-4(?-ene (HSC-8D-P8), thymidine (HSC-8E-P1), 2-deoxyuridine(HSC-8E-P2), were isolated and identified from the soft coral Dendronephthya sp.
