Biochemical assay is very important in life .science, clinical diagnosis, and food quantative control. Electro-analytical method is very sensitive, selective with simple instrument and it is seldom disturbed by the color or turbid of biological samples. So bioelectroanalytical chemistry is the research focus of recent years.The thesis is supported by the National Natural Science Foundation of China (Grant NO. 29775012, NO. 20075013) and Science & Technical Foundation of Qingdao City (Grant NO. 2000-GS-2-02). In this thesis, the electroanalytical method was combined with enzymatic assay, immunoassay and protein assay to establish a new detection method for alkaline phosphatase, horseradish peroxidase and proteins respectively. The established methods were applied to the HRP-labeled enzyme-linked immunoassay for some important plant viruses and to the determination of content of alkaline phosphatase, serum albumin in healthy human serum. Compared with the spectrophotometric method, the results are satisfactory and reasonable. The primary contributions of this research are:1) New electrochemical methods for the determination of alkaline phosphatase were established based on the p-nitrophenyl phosphate, phenyl phosphate, 2-ascribic acid phosphate as substrates and differential pulse voltammetry as the detection method, respectively. The established methods were applied to the determination of alkaline phosphatase in healthy human serum and the results were in accordance with the spectrophotometric method.2) New voltammetric enzyme-linked immunoassay systems were established using p-phenylenediamine, o-phenylenediamine, p-aminophenol, o-aminophenol and 3, 3', 5, 5'-tetramethyl benzidine as substrates.
