In this paper a sensitive electrochemical enzyme-linked immunoassay was proposed for the detection of detin sialophosphoprotein by using horseradish peroxidase(HRP) as the labeled enzyme. By using the3,3’,5,5’-Tetramethylbenzic (TMB), o-tilidine(OT), o-phenylenediamine(OPD), o-dianisidine(ODA) reaction as the substrate, the enzymatic catalytic reaction product can be electrochemical detected by the second order derivative linear sweep polarography with high sensitivity, which was exhibited a sensitive second order derivative linear sweep polarographic reduction peak. This electrochemical detection of DSPP was further coupled with a sandwich ELISA procedure for the detection of DSPP. DSPP is an important marker of orthodontic root resorption. So a new sensitive method of the electrochemical detection of trace DSPP could be established.This paper report that carboxyl-modified graphene oxide(GO-COOH) has Peroxidase-like activitythat can catalyze the reaction of peroxidase substrate3,3,5,5-tetramethylbenzidine(TMB) in the presence of H2O2to produce a blue color reaction. The generated product exhibited a sensitive second order derivative linear sweep voltammetric reduction peak in Britton-Robinson buffer. GO-COOH has even higher catalytic activity to TMB than the natural enzyme horseradish peroxidase(HRP). This proposed method was further applied to determine the H2O2content in fresh milk samples with satisfactory results.
