| Objective: To study the craft of acanthopanacis senticosi astragali gelatum preparation and to investigate the effects of the gelatum on human sperm motility and mobility of asthenospermia in vitro.Methods: We extract the main component from crude traditional Chinese medicine acanthopanacis senticosi and astragali by water decoction and alcohol precipitation. The gelatum is prepared with carbomer-940 as base material and we investigate quality control of preparative craft. By using computer-assisted sperm analysis (CASA), we have observed the effects of the gelatum on human sperm mobility in vitro. The sperm that obtained by masturbation from 50 men with asthenospermia were incubated with acanthopanacis senticosi astragali gelatum. Measurement was carried out at 15, 30 and 60 minutes in all specimens.Results: The performulation is reasonable and preparation craft is feasible. PH of the gelatum is 6.5～7.5, stability is good and it has no cutaneous stimulation. Acanthopanacis senticosi astragali gelatum can increase human sperm mobility of asthenospermia. Compared with control group, the difference of percentage of viability (Mot), percentage of progressive mobile sperm (A+B%), the curvilinear velocity (VCL), the straight line velocity (VSL) and average path velocity (VAP) is distinct (P＜0.05). Conclusion: Preparative craft of acanthopanacis senticosi astragali gelatum is convenient and fit for hospital production and clinical use. It is a kind of valid drug therapy for asthenospermia.
