| There are three parts in this dissertation. Studies on isolation and characterization of two glucan and five heteropolysaccharides from Cynanchum panilatum (Bunge) kitagawa were discussed in the first part. Studies on relationship between chemical modification and biological activities of lentinan were discussed in the second part. The third part mainly concerned on studies on method of methylation analysis.1. 1 Studies on glucan I from. Cynanchum panilatum (Bunge) kitagawaA homogenous glucan, CPB-1 was isolated from Cynanchum panilatum (Bunge) kitagawa. Its means molecular weight was estimated to be 1.5×10~4 from the standard curve with Dextran T system determined by HPLC method, and its specific ratation was +159.2°. Sugar analysis showed this polysaccharide was only composed of glucose. Methylatin analysis showed it was mainly composed of 1,4 linked glucosyl residue. Partial hydrolysis and acetolysis indicated that its side chain was composed of 1,4 and 1,6 linked glucosyl residue, attached to the 0-6 of main chain with 1,4 linked glucosyl residue. The a configuration is confirmed by IR and NMR data. Compared with similar glucan, this polysaccharide owns 1,6 linked glucosyl residue which didn't appear in the other glucan, showing it is a glucan with new structural feature.1.2 Studies on glucan II from Cynanchum panilatum (Bunge) kitagawaThis polysaccharide was purified from the second fraction eluted with 0.1 mol/L NaCl solution on DEAE-Sepharose FF column followed by Sephadex G-75 column. Sugar analysis showed CPB-2 was composed of glucose.
