"Dong Qin"Xiangju Pian produced by Shaanxi Xiangju pharmaceutical group with Platycarya Strobilacea Sieb.et Zucc.as raw materials has good results in the treatment of rhinitis and sinusitis.In condition that the production process is still without quality standards and has difficulty in the production process control.There are a lot of obstacles in the product quality upgrading process.To meet the urgent need for the actual production,a series of studies on active ingredients from Platycarya Strobilacea Sieb.et Zucc containing extraction,purification,related dynamics and in vitro antibacterial activity are carried out.This paper,an integral part of the studies,mainly achieves the following results:(1) An ideal extraction process and an optimum operation condition which are used in the route to extract polyphenol and flavonoids from Platycarya Strobilacea Sieb.et Zucc are obtained.The optimum polyphenol extraction are as follows:temperature of 77℃,alcohol volume fraction of 55%,the ratio of solvent and material 14 mL/g,extracting 3 times and extraction time of 1.5h.The extraction rate of total polyphenol could be up to 3.646%.While, the optimum flavonoids extraction is under the condition with temperature of 60℃,alcohol volume fraction of 65%,the ratio of solvent and material 9 mL/g,extracting 3 times and extraction time of 2h.The extraction rate of total flavonoids could be up to 4.29%.The test has good repeatability comparison with the pilot inspection and gets a higher extraction rate by 1%than that of the existing level of production enterprises.(2) Establishing the kinetic model in extracting the active ingredients from Platycarva Strobilacea Sieb.et Zucc.and obtaining the polyphenol and flavonoids rate constant of extraction,residual rate of extraction,activation energy,half-life,a series of effective diffusion coefficient of the kinetic parameters.The results provide the theoretical basis and reference for the design and optimization of extraction conditions used in the active ingredients from Platycarya Strobilacea Sieb.et Zucc.(3) Macroporous resin is a suitable route to purify polyphenol and flavonoids from Platycarya Strobilacea Sieb.et Zucc.Through static adsorption,desorption,dynamic adsorption,desorption experiments,relative parameters of adsorption kinetics and thermodynamics are acknowledged.The result is shown thatL-327F and H-327B are ideal resins separately for purification of polyphenol and flavonoids.Optimization of polyphenol purification conditions are as follows.Adsorption process:concentration of sample solution 4.9mg/mL,with a flow rate of 2BV/h.Elution process:using 70%of ethanol and 4BV ethanol as eluant.The purity of polyphenol reaches 56.032%after purification by macroporous resin. While flavonoids adsorption and elution process are that concentration of sample solution is 8.9mg/mL,with a flow rate of 3BV/h,use 90%of ethanol and 5BV ethanol as eluant.The purity of flavonoids reaches 62.72%after purification.(4) Two monomer compounds are firstly separated from Platycarya Strobilacea Sieb.et Zucc,which are classified as gallic acid and quercetin after determination of physical and chemical properties as well as spectrum analysis.(5) After in vitro antibacterial experiments of kinds of representative bacteria, Staphylococcus aureus,Escherichia coli,Streptococcus pneumoniae and Pseudomonas aeruginosa,which are mainly bacteria infection of sinusitis,rhinitis,it shows that polyphenol and flavonoids extracted from Platycarya Strobilacea Sieb.et Zucc are of the main active ingredients.Both of them have strong inhibition.The results of this study can help the pharmaceutical group to improve production technology and product quality,establish product quality standards,and accelerate the development of second-generation products.It has important practical value and theoretical significance to Shaanxi Xiangju pharmaceutical group and some reference value on similar studies.
