Platycarya Strobilacea Sieb.et Zucc belongs to Platycarya and Juglandaceae.Its infructescence has been used as the main drug for treating nasal sinusitis and acute or chronic rhinitis.Plant polyphenols are widely applied to pharmaceutieal,food and cosmetic industries,since they contain a variety of biological activities,such as anti-oxidationanti-free radicals, anti-ultraviolet and so on.The present studies focus on the technology of extraction,separation and purification and the determination of total content of polyphenol from infructescence as well as antibacterial activity.The rearch established a set of methods, which are economic and safe,and are suitable to industrial produetion.The main studies are as follows:(1)The study established an ideal method for the determination of total content of polyphenol from infructescence vir UV with ferrous tartrate as reagent.The method has the particularity of simple operation,high accuracy,sufficient stability,and good reproducibility.(2)In the extraction experiment polyphenol is extracted by ethanol reflux extraction.At first the conditions for the extraction were studied by single-factor test, then the conditions were optimized by Response Surface Methodology.The optimum conditions are as follows:extracting with 55% acetone solution,temperature at 77℃, extracting 3 times and every time 1.5h with ratio of raw materials and solution was 1g:14mL.(3)With the recovery rate of polyphenol as indexes,optimum process conditions for separation and purification of polyphenol from infructescence of Platycarya Strobilacea Sieb.et Zucc with macroporous resin was studied. By static and dynamic adsorption and desorption, L-327F type macroporous resin is the best for separation and purification of polyphenol among seven types of macroporous adsorption resin chosen. The optimum process condition were concentration of feed 4.9mg.mL, feeding rate 2BV/h,and eluted with 4BV of 70% ethanol.Then studied the theoretical about adsorption dynamics and thermodynamics of L-327F type macroporous resin.(4) One component was separated by means of silica gel and chromatography on Toyopear and Sephadex LH-20.The structures were determined by spectral analysis and chemical evidences.And it is identified that the component is Gallic acid.(5) In the research the inhibition zone, the minimum inhibitory concentration, minimum bactericidal concentrations of plant polyphenols were measured. And the results showed that the polyphenols from infructescence of Platycarya Strobilacea Sieb.et Zucc have a strong bacteriostasis effect.
