Study On Identification Of Hybrids And Hereditary Variation Of Natural Pollinated Chinese Jujube Seedlings

Two groups of Dongzao natural pollinated seedlings were used to identify hybrids from Dongzao x Jinsixiaozao, Dongzao x Jianzao and Dongzao x Linyilizao with AFLP technology. The segregation pattern of AFLP markers in hybrids from Dongzao x Linyilizao was studied, and a genetic map for Chinese jujube was constructed. The segregation pattern of leaf peroxidase isozyme of Dongzao seedlings was studied. Trait segregation of hybrids from Dongzao x Jinsixiaozao, hybrids from Dongzao x Jianzao, and natural pollinated seedlings of 20 varieties of Chinese jujube as female parent was studied.(1) Hybrids in Huanghuadongzao seedlings and Renxiandongzao were identificated with AFLP technology. 12 pairs of primers were found to be highly polymorphic and used in the AFLP analysis of 65 Huanghuadongzao seedlings. Altogether 517 bands were produced among which 376 bands were polymorphic ones with the percentage of polymorphism being 72.7%. 10 markers were present only in Jinsixiaozao, and 34 hybrids from Dongzao Jinsixiaozao were identificated. 7 markers were present only in Jianzao, and 15 hybrids from Dongzao x Jianzao were identificated. 7 pairs of primers were used in the AFLP analysis of 85 Renxiandongzao seedlings. 22 markers were present only in Linyilizao, and 72 hybrids from Dongzao x Linyilizao were identificated.(2) An F1 population from Dongzao Linyilizao was used to study the segregation pattern of AFLP markers and construct a genetic map for Chinese jujube. The polymorphism of AFLP markers in the F1 population from Dongzao x Linyilizao was high, and the frequency of the segregation loci was 33%. The segregation patterns included Mendelian segregationmodel and deviation from Mendelian segregation model. Mendelian segregation models included non-segregation, segregation ratio of nearly 1:1, and segregation ratio of nearly 3:1. The frequency of non-segregation, segregation ratio of nearly 1:1 and 3:1, and deviation from Mendelian segregation were, respectively, 67%, 23.8%, 9.2% of 294 AFLP markers. 28 AFLP markers were assigned to 7 linkage groups covering 458.66 centimorgan (cM) of Chinese jujube. The average interval beween these markers was 16.38 cM.(3) Leaf peroxidase of Huanghuadongzao seedlings was studied. Four zymogram types of leaf peroxidase of Dongzao seedlings were obtained according to the appearance of isozyme band. The frequency of zymogram type I,II, HI, IV were 31.2%, 54.8%, 5.4%, 8.6% respectively. The heredity conduct of the single isoperoxidase band conformed to Mendelian law of segregation. It was proved that peroxidase isozyme was controled by a single gene. According to the heredity conduct of the isoperoxidase POD2 band, it was proposed that Jinsixiaozao and Jianzao could be dominant heterozygote, and Dongzao could be recessive homozygote.(4) The trait segregation of branch, leaf, spine, characters of 34 hybrids from DongzaoxJinsixiaozao and 15 Chinese jujube hybrids from Dongzao * Jianzao were studied. The results showed that the plant height, diameter at butt-end, number of branch, length between nodes on the trunk, Leaf length, Leaf width and length of spine of hybrids from DongzaoxJinsixiaozao presented obvious segregation, and their variance coefficient was more than 20% respectively. Variance coefficient of the plant height, number of branch, Leaf width, length of spine of hybrids from Dongzaox Jianzao was more than 20% respectively. Variance coefficient of diameter at butt-end, length between nodes on the trunk, Leaf length, Leaf width, leaf index and length of spine of hybrids from DongzaoxJinsixiaozao were more than that of hybrids from Dongzao Jianzao. Color of vegetative branch, leaf shape and color of leaf of Dongzao hybrids displayed the variation in various degrees besides displaying parents properties.
