Cow endometritis is common obstetrics in puerperal disease for bacterial infection, which causes large economic losses. The study included 12 cows with normal puerperium and 42 cows with endometritis on different farms in Heilongjiang province.All of bacteria in uterus secretions were cultured in anaerobic and aerobic conditions.8 bacteria were isolated in normal cows and 14 bacteria were isolated in cows with endometritis.The dominant pathogen in cows with endometritis were Escerichia coli, Staphylococcus, Arcanobacterinm, Bacteroides and Fusobacterium.In order to provide an important evidence on bacteriostasis for clinical bovine endometritis, the experiment in vitro was done for 16 kinds of antibiotics,11 kinds of the Chinese herbs and on inhibiting the pathogen bacteria of dairy cow endometritis. The result indicated that the pathogen bacteria were markedly inhabited by vancomycin, neomycin, florfenicol, oxybicycline and ciprofloxacin, especially the ciprofloxacin had better effect; Sophora flavescens, scutellaria, heartleaf houttuynia herb, Dandelion, Brunellae spica and two compositions greatly inhibited the pathogenic bacteria's growth.The composed chinese herbal, which is used for therapy on endometritis in dairy cow, were screened out by orthogonal principle. Firstly, bacteriostatic test in vitro were done on dominant pathogen in cows with endometritis. Then, three anti- inflammatory tests in vitro were done. Finally, the experiment on eye irritation and skin anaphylaxis were done respectively in rabbits and cavies. The result indicated that the components of chinese herbal, which is made up of the Dandelion, Forsythia, Honeysuckle and other 5 kinds of Chinese herb, had apparently bacteriostatic functions ,had markedly anti-inflammatory, had no anaphylaxis and had gentle irritation on mucosa system.Twenty-four Friesian cows with endometritits were used in this study. The dairy cows were randomly assigned to four groups: in control group (Group-Ⅰ,n=5), only 30ml sterile PBS was infused, Group-Ⅱ(n=5)were treated with the composed chinese herbal, Group-Ⅲ(n=9) were treated with E.coli LPS(100μg), Group-Ⅳ(n=5)were treated with E.coli LPS and compositions. Intrauterine application Escherichia coli lipopolysaccharide (E.coli LPS)as a uterine defense stimulator was used in cows with bacterial endometritis.Uterine washing fluids showed significant increase in the total leucocytic count,percent of neutrophils and alive neutrophils in treated Group with E.coli LPS. There were no obvious changes about all value in control group(group-Ⅰ). In the treatment group-Ⅱ, the content of IL-2, IL-6, IL-8, TNF, MDA and T-AOC in serum had no obvious change, the activities of GSH-Px,SOD in serum significantly increased in 48h(P<0.05).In the treatment group-Ⅲ, the content of IL-2,IL-6 and TNF in serum had no obvious change, the values of GSH-Px,SOD,MDA and T-AOC in serum had no obvious change, the content of IL-8 in serum increased in 48h and then decreased to normal levels. In the treatment group-Ⅳ, the values of IL-2,IL-6,IL-8,TNF,GSH-Px,SOD,MDA and T-AOC in serum had no obvious change, but the total leucocytic count,percent of neutrophils and alive neutrophils increased more obvious than in treated Group with E.coli LPS. It was conclude that administration of Intrauterine E.coli LPS stimulated uterus defense mechanisms and cleared the infection in uterus.Three of the 5 cows conceived in the group-Ⅱ, six of the 9 cows conceived in the group-Ⅲ, four of the 5 cows conceived in the group-Ⅳwhile only one cow conceived in control group(group-Ⅰ).
