Cage layer osteoporosis is defined as a progressive loss of structural bone, leading to increased fragility and susceptibility to fracture during the laying period. Husbandry, nutrition, hormones and genetic factors may play some roles in forming of osteoporosis, but the exact mechanism is unknown. Bone metabolism development of ISA layers was studied, and effects of Gushukang on controling osteoporosis in cage layers and its mechanism were discussed in this paper.1 Age-related Changes of Bone Metabolism in ISA LayersChanges of bone metabolism in ISA layers with 15, 35, 58 and 70 week-age were studied. The results were as follows: Compared with the 15 week-age group, AKP and TRAP levels in 70 week-age group decreased 61.06% and 54.38%(P<0.05), respectively, the radiographic density of humerus, tibia, femur and keel increased 35.23%, 34.86%, 28.38%and 34.86%(P<0.05), respectively. Compared with the 15week-age group, cortical width of humerus, tibia and femur in 70 week-age group decreased 11.43%, 36.96% and 28.75%(P<0.05), respectively, percentage of cortical volume decreased 13.97%, 38.02% and 28.53%(P<0.05), respectively. In Conclusion, the bone formation was decreased while its resorption was increased in 70 weeks-age ISA layers with severe osteoporosis. It is one of important factors of pathogenesis that continued resorption of cortical bone leads to osteoporosis throughout the laying period.2 Effects of Gushukang on Bone Metabolism of Cage LayersEffects of Gushukang (GSK) on bone metabolism of ISA layers were studied. 90 ISA layers of 58-week old were randomly assigned to three groups. The control was provided with basal diet, the GSK group was provided with basal diet with addition of 1% GSK, the Ipriflavone (IP) group was provided with addition 25 mg/ Kg Ip. The experiments of three groups lasted for 90 days. The results indicated that the levers of Ca, P, AKP, TRAP, BGP in the serum were lower significantly, bone index, bone radiographic density, cortical width, cortical area and cortical area ration of humerus, tibia and femur, and the lever of serum IGF-I were higher significantly in the GSK-group and IP- group than the control. There was not significant difference between GSK-group and IP-group. It was revealed that GSK can accelerate bone formation and inhibit bone resorption and improve bone metabolic profile of ISA layers during the later laying period.3 Effects of Gushukang on Production Performance, Egg Quality and Bone Metabolism of Cage Layers in Intensive FarmsEffects of Gushukang (GSK) on bone metabolism of ISA layers were studied. 1000 ISA layers of 55-week old were randomly assigned to two groups. The control was provided with basal diet, the GSK group was provided with basal diet with addition of 1% GSK. The experiment lasted for 10 weeks. The results indicated that broken eggs and feed conversion ratio were decreased significantly, laying ratio and bone index, bone radiographic density, cortical bone index, cortical bone area ratio of humerus, tibia and femur were increased significantly, bone strength of tibia also was increased significantly, medullary bone area ratio of tibia and the lever of serum AKP were decreased significantly in the GSK-group than the control. It was revealed that GSK could increase production performance, egg quality, improve bone metabolism and control osteoporosis in cage layers during the later laying period.4 Effects of Gushukang on Proliferation and Activity of OsteoblastsEffects of Gushukang serum on the proliferation and activity of osteoblasts were studied. Obtained from the chicken calvariae by sequential enymy digestion, the osteoblasts were cultured with different concentration serum of Gushukang. MTT and PNPP methods were made to assay the proliferation and activity of osteoblasts. The results showed that Gushukang serum stimulated the proliferation of osteoblasts and activity of AKP from osteoblasts, compared with the control. The effects of the 5 h- group of Gushukang serum was the best among the other groups. It was revealed that Gushukang could stimulate proliferation, differentiation and activity of osteoblasts, and accelerate bone formation.5 Effects of Efficacious Components in Gushukang on Proliferation and AKP Activity of OsteoblastsEffects of efficacious components in Gushukang on the proliferation and AKP activity of osteoblasts were studied in this experiment. The osteoblasts cultured with Naringin and Astragaloside of different concentration, MTT and PNPP methods were used to assay the proliferation and AKP activity of osteoblasts after 24 h, 48 h and 72 h. The results showed that Naringin and Astragaloside at concentration from 10 to 80 ng/mL stimulated the proliferation of osteoblasts and activity of AKP. Effects of concentration at 40 ng/mL was the best than the others. It revealed that effects of Naringin and Astragaloside could stimulate proliferation, differentiation and activity of osteoblasts, and accelerate bone formation.
