| Tilia miqueliana is deciduous tree of Tiliaceae.It can be used as timber tree,honey plant and ornamental plant.Based on T.miqueliana wild resources investigating,Community enology,population ecology, the individual growth disciplinarian and biomass of it were studied.The genetic diversity of T. miqueliana populations was analyzed by ISSR and RAPD molecular marks.In order to utilize and protect this species,tissue culture was also studied.The results were as follows:1.The wild resource of T.miqueliana is rare.Results of the investigation showed that they grow in forests in the vicinity of temples,and rare in the form of woods.The smallest population only has about 10 stubs where as the biggest population has several hundred stubs.They distribute in dot shape and block shape.2.Statistically,there are 114 species of vascular plants that belong to 55 families 95 genera inter-dependent on T.miqueliana.In the context of the tree layer,the significant value of the T. miqueliana is higher than other species,which indicates that it has an absolutely advantage in the community.T.miqueliana has a broad range of niches,showing that the specialization of T. miqueliana is weak,and it has a good adaptability to all kinds of habitats.Moreover,viewing the overlap value of every niche,most of them are more than 0.03,explaining that in the community every species has an obvious trend of resources sharing,and they compete with each other.3.The T.miqueliana population's age structure basically all assumes the inverted pyramid shape, the T.miqueliana population is the decline population,the seedling quantity of the population quite few,but the big tree are many;The population's survival curve basically belongs to DeeveyⅢ;In the population of T.miqueliana,the mortality rate of the seedling is big,the habitat of the seedlings is restricted,the mortality rate is high,the number of the juvenile age seedlings are few,the renewal was poor.The population assumes the declining characteristics basically.4.Morphological analysis showed that the greatest variation of T.miqueliana is its bract stalk's long.The variations of its long leaf stalk,long seed stalk and bract are also great.The seed variation index is the smallest,and the properties of seeds are the most stable.The mean variation index of the population in Mt Baohua,Jiangsu is the biggest,which showed that the morphology of this population is rich.This meets the results of molecular marker.5.ISSR and RAPD molecular marker tested the diversity of the 157 trees in 8 population communities.The respectively selective 11 and 10 primers detected 80 and 169 loci, respectively,among which there are 77 and 153 polymorphic loci respectively,and the percentage of them reached 96.25%and 90.58%respectively.ISSR and RAPD molecular marker test results showed that in the T.miqueliana species level,among the diversity indexes, the Shannon 0.4171and 0.3652,the gene diversity 0.2706 and 0.2304,and effective number of alleles(Ne)is 1.4463 and 1.3604;and by the genetic variation analysis the gene differentiation quotiety 0.5061 and 0.4353,community gene flow 0.4879 and 0.6487,and average value of population community genetic gap0.2070 and 0.1468 respectively.Result show that①The genetic diversity of Mt.Baohua population community is the richest and Huangzangyu is the next.②T.miqueliana genetic variation mainly existed among the population community and was not very obvious.③The result of UPGMA was not consistent with the geographical distribution,and never reached a remarkable level of every environmental factor.It is because T. miqueliana wild population community is naturally renewed after planted,and not the real primary community.④The genetic differentiation quotiety of T.miqueliana is 0.43-0.51,in order to preserve its genetic diversity such population communities of higher genetic diversity as Anhuihuangzangyu and Jiangsu Mt.baohua must be saved.6.Analysis result of timber showed that T.miqueliana in Huangzangyu grow fastest,especially the mean growth velocity in height(0.35m per year)and the timber mean growth volume (0.0113 m3),which showed that the T.miqueliana is suitable to grow in this environmental condition.T.miqueliana's timber volume maturity age is between 50 to 70 years old,from the perspective of timber wood;the best cutting age of T.miqueliana is 50-70 years old.The T. miqueliana's maximum diameter,the average tree height and the continually annual growth volume basically appeared between 6 to18 years old,which explained that its earlier period growth speed is comparatively faster.7.The biomass of T.miqueliana was studied by using direct gain method.The result showed that it is of low biomass,and great differences existed among individuals and population communities,and of T.miqueliana,truck biomass>root biomass>branch biomass>leaf biomass.8.In the system of organized cultivation propagation,the best explants material is stem tip and axillary bud of the one year seed tree.The best culture medium combination of bud differentiation is:MS+6-BA0.5mg/L+NAA0.02mg/L+GA3 1.0~2.0mg/L,the best culture medium combination of multiplication is:MS+BA0.3+IBA0.02+GA30.3,and the best culture medium combination of rooting is:MS+BA0.05mg/L+IBA1.0 mg/L.The rooting plant will be gained from this system.9.The T.miqueliana's is of obvious great ornamental value,having the basic characteristics of the ornamental plants.It is an ornamental integration of the tree shape,the tree posture,the trunk,the leaf,the flower and the inflorescence.It also served as the roadside trees,the courtyard shade tree,main trees of residential area evanescence and so on.It can plant in patterns such as isolated planting,group planting,scatter plot planting,or in the form of the high hedge. |
