Tilia miqueliana is a multi-functional tree species with ornamental,greening,timber,medicinal and edible value.In this study,through three consecutive years,plus trees were selected from the natural forests of T.miqueliana distributed in Anhui Huangcangyu Nature Park and Temple Generous Provincial nature reserves.The plus trees with high ornamental and greening values were selected,which laid a foundation for the breeding of fine varieties with high ornamental values.For the first time,the comprehensive evaluation system of ornamental characteristics of T.miqueliana was established by analytic hierarchy process(AHP).In order to further improve the breeding of T.miqueliana and provide excellent regional materials of a single clone,this study discussed the establishment of rapid propagation system of T.miqueliana with bud and stem as explants.The main results were as follows:1.Based on the natural forest tracing of T.miqueliana as the dominant species in Suzhou City,Thirty plus tree were selected,and eight factors including tree growth,pest resistance,physiological indicators and shape indicators were selected.Making a observation of the phenological period of the primary tree species,and a comprehensive evaluation system of landscape effect application value was established by applying the analytic hierarchy process(AHP),that is,grading its crown width,tree height,breast diameter,tree posture,height under branches,leaf density,leaf color and other characteristics step by step,and calculating the weight of each character according to the breeding objective,adding the product of each index score and weight.The higher the total score was selected,to determine the best fine individual 3 are N11,N21,N26,respectively,which was characterized by dry concept shaped,green,foliage species.2.In preparation for the regional test varieties to explore T.miqueliana plant tissue culture techniques.The stems with buds of the superior single plant were used as explants,using single factor experiment design to explore the best culture conditions for the adventitious bud differentiation,proliferation and rooting of T.miqueliana.And the callus was induced by different disinfection methods,sampling time,medium type and pH value of media.The results showed that the best disinfection method was the 60s with 75%alcohol+8min 0.1%HgCl2,which could reduce the pollution rate to 16.67%and the survival rate up 64.44%.The best sampling time was in April,when the pollution rate of explants was the lowest of 8.3%,and the survival rate could reach 80%.The best medium type was MS medium,and pH 5.8 was the best pH value.The most suitable medium for adventitious bud differentiation was MS+BA 1.0 mg/L+NAA 0.1 mg/L+KT0.05 mg/L(pH 5.8).The differentiation rate was 83.33%.The optimum medium for bud proliferation and subculture was MS+0.3 mg/L 6-BA+0.03 mg/L IBA+0.5 mg/L GA3,and its multiplication coefficient could reach 5.34.The optimum medium for rooting was 1/2MS+6-BA0.1 mg/L+IBA1.0 mg/L+5.0 g/LAC,and the rooting rate reached 56.6%after one month.After the domestication of rooting seedlings,transplanting in the substrate(nutrient perlite:vermiculite:soil=1:1:2mixed),the survival rate reached 55%high.
