PCR-SSCP and PCR-RFLP method was used to study the SNP of Apn (Adiponectin) Gene, MDH (Malate Dehydrogenase)Gene and IGF-â… (Insulin-like growth factor-â… ) Gene that might be correlated with fat deposit traits in Jinghai Yellow chicken, and the systematic investigation was conducted on Jinghai Yellow chicken Apn Gene, MD HGene and IGF-â… genes to evaluate the association of molecular markers with growth traits, reproduction traits, carcass traits and meat quality traits. The main results are listed as follows:1. Parts of 5'-flanking region and whole code region DNA of Apn gene were obtained using the techniques of PCR-SSCP.7 Apn SNPs were found in Jinghai Yelow chicken:C1251G, C1608T, G1773A, A1782G, T1902C, G1993C and G2013A, and they were all synonymous SNP. A single mutation at the position of the 235 bp of MD 5'-flanking region was found. A SNP of IGF-â… was detected.2. AD2,AD7,MD and P11 locus have medium level PIC value, AD5 and AD6 have high level PIC value.3. At the AD2 locus of Apn gene, birds with AA genotype has significant lower value of age at first egg than the birds with AB genotype (P<0.05), birds with AA genotype has significant higher value of egg numbers at 300 days than the birds with AB genotype (P<0.05).At the AD5 locus of Apn gene, birds with h3h5 genotype has significant lower value of age at first egg than the birds with h1h1 genotype (P<0.05), birds with h3h5 genotype has significant higher value of egg numbers at 300 days and 66 weeks than the birds with h1h1 genotype (P<0.05). At the AD6 locus of Apn gene, h2h5 genotype was the predominant genotype that increased egg numbers.At the AD7 locus of Apn gene, birds with GG genotype has significant lower value of age at first egg than the birds with AA genotype (P<0.05), birds with GG genotype has significant higher value of egg numbers at 300 days and 66 weeks than the birds with AA genotype (P<0.05). Major effect index (MEI) of reproduction traits was low; it implied that Apn gene could be linked to a major gene to significantly affect reproduction traits in chicken4. AD2,AD5 and AD6 locus of Apn gene have significant effect on abdominal fat deposition traits:Chickens with BB genotype at AD2 locus have higher value of abdominal fat weight and abdominal fat ratio than birds with AA genotype (P<0.05) in male population, but the value of abdominal fat ratio of BB genotype has higher value than the birds with AA in female birds(P<0.05). CP of abdominal fat ratio of BB genotype was over 30%.Birds with hlh3 genotype at AD5 locus have significant higher value of abdominal fat weight and abdominal fat ratio than birds with h2h3 genotype (P<0.01) in female population, and CP of abdominal fat weight and abdominal fat ratio of hlh3 genotype were 48.71% and 12.72% respectively.Birds with h3h4 and h2h5 genotype at AD6 locus have significant higher value of abdominal fat weight and abdominal fat ratio than birds with h2h3 genotype (P<0.05) in female population, and CP of abdominal fat weight and abdominal fat ratio of h3h4 genotype were 40.98% and 10.99%, and for h2h5 were 18.68% and 32.93%, respectively.5. Significant differences were found in egg numbers at 300 days and 66 weeks between birds with BB genotype in MD locus and the birds with AA and AB genotype(P<0.01). Chickens with BB genotype have lower value of abdominal fat weight and abdominal fat ratio than birds with AA genotype (P<0.05). BB genotype was the predominant genotype that decreased abdominal fat, CP of abdominal fat weight and abdominal fat ratios of BB genotype were -24.18% and -30.18% respectively in males and -33.65% and -18.24% respectively in females. It implied that Apn gene could be the major gene or is linked to the major gene that affects the growth and reproduction traits in chicken.6. Significant difference was found in talon weight between the chickens with DD genotype and the birds with CC genotype of IGF-â… . CP of talon weight of CC and DD genotype was over 11% in both sexes.7. Significant or very significant interaction effect of Apn, MDH and IGF-â… were found on birth weight. Significant interaction effect of Apn and IGF-â… were found on day of first egg and eggs at 300 day in Jinghai Yellow chicken...
