In this study,Jinghai Yellow Chickens were used as experimental materials,the IL-6,IL-8(CXCLi2)and CCLi2 mRNA expression in different tissues were determined by relative quantitative RT-PCR.After that,DNA sequencing technology was used to detect the SNPs of IL-6(interleukin 6)gene in Jinghai Yellow chickens.The general linear model was used to analyze the relationship between SNPs and coccidiosis-resistant parameters.The main results were described as follows:1.The IL-6 gene was expressed in all tissues.In tthe infection group,we detected the expression of IL-6 in the tissues of cecum,thymus and kidney were significantly higher than that in the same tissues of control group(P<0.01),and the IL-6 expression of the infection group in bursa was significantly higher than that in control group(P<0.05).The IL-8 gene was expressed in all tissues.The IL-8 expression of the infection group in the glandular stomach,cecum and thymus was significantly higher than in control group(P<0.01),while it was significantly higher in the spleen and small intestine than in control group(P<0.05).The CCLi2 gene was expressed in all tissues.The CCLi2 expression of the infection group in the spleen and thymus was significantly higher than in control group(P<0.05),while it was significantly higher in the glandular stomach,cecum and kidney than in control group(P<0.05).2.We choose IL-6 as a candidate gene,and found 28 mutations in the sequence of IL-6.Compared with the GenBank,we found four new mutations.19 SNPs were located in the 5'-upstream regulation region,removing the GenBank recorded SNPs,three mutations were newly discovered;2 SNPs were located in the introns region;2 SNPs were located in the exon region,both of them were synonymous mutations;5 SNPs were located in the 3'-upstream region.3.Tthe general linear model was used to analyze those SNPs's relationship with coccidiosis-resistant parameters.We found that 13 SNPs were associated with resistance index.N0.1,NO.6 and N0.25 SNPs were significantly associated with MDA;N0.7,NO.12,N0.16,N0.17,NO.20 and NO.21 SNPs were significantly associated with IL-2;NO.12,NO.15 and NO.17 SNPs were significantly associated with GSH-Px;NO,14 SNP was significantly associated with NO;NO.17 and NO.19 SNPs were significantly associated with IL-17;NO.19 and N0.28 SNPs were significantly associated with CAT;NO.19 SNP was significantly associated with IL-16.4.Compared with the concentration of IL-17 in different haploid type combinations,H2H3 was significantly higher than other haploid type combinations(P<0.05);compared with the concentration of GSH-Px,H2H5 was significantly higher than H1H1,H1H2 and H1H5(P<0.05);compared with the concentration of MDA,H1H2,H1H4,H2H5 were significantly higher than H1H1,H1H5,H2H3(P<0.05);compared with the concentration of IL-2,H2H5 was significantly higher than H1H1,H1H2 and HlH5.The NO concentration of H2,H3' was significantly lower than H1'H3'(P<0,05),while it's CAT concentration was significantly lower than H2'H2'(P<0.05);compared with the concentration of ISOD,H2'H2'was significantly higher than H2'H3',H3'H3'(P<0.05),the H1'H5' was significantly higher than H3'H3'(P<0.05);The IL-2 concentration of Hl'H2',H2'H3' were significantly higher than H1'H1'(P<0.05),while the IL-16 concentration of them were significantly higher than H1'H3'?H1'H5'(P<0.05).
