The Effects Of The "Xingnao Kaiqiao" Acupuncture Method On Cytokine And Adhesion Molecule In Focal Cerebral Ischemic Rats

Objective:To observe the inflammatory morphologic changes and it's related changes of cytokine and adhesion molecule in focal cerebral ischemic rat,and also the effects of the "Xingnao Kaiqiao" acupuncture method on them.Method.(l) In this study we used one -side middle cerebral artery occlusion rats as focal cerebral ischemia experimental animal,which were made by thermocagulation method. (2) Use morphologic method to observe the inflammatory reaction in ischemic brain, especially the phenomena of adhesiveness and infiltration of the inflammatory cell.(3) Use radioimmunoassay method to assay the levels of the IL-1 P , EL-6. IL-8, and TNF- a in the brain tissues and the blood serum.(4) Use the immunohistochemical and the in situ hybridization method to observed the expressions of the TNF-a , E-selectin P-selectin ICAM-1 TGF-P 1 and their mRNA in ischemic brain tissues. Resultsr(l) After ischemia,the inflammatory cell adhered to the vascular endotheliocyte and infiltrated to the brain tissues around the ischemic area,and this phenomenon aggravated as the ischemia continuing.After treating by acupuncture,the necrosis areas reduced and and the degree of the adhesiveness and the infiltration of the inflammatory cell also decreased.(2) The levels of the IL-1 P IL-6, IL-8 TNF- a in the brain tissues and the levels of the IL-8, TNF-a in the blood serum increased significantly after ischemia, the accupuncrure may decrease their levels significantly.the level of IL-6 in blood serum increased in 3 hours after ischemia,and retured to normal range from 24 to 48 hours after ischemia.Accupuncrure can decreased the level of the IL-6 in blood serum during the early ischemic period(from 3 to 12 hours),and increased its levels during the later ischemic period (from 24 to 48 hours).(3) The expression levels of the TNF- a ,E-selectin,P-selectin,ICAM-l, TGF- P 1 and their mRNA increased significantly after ischemia.Acupuncture can decrease the expression of TNF- a ,E-selectin,P-selectin,ICAM-l and their mRNA ,and increase the expression of TGF- P 1 and its mRNA significantly. Conclusion:" Xingnao Kaiqiao " acupuncture method can reduce the inflammatory reaction and protect the ischemic brain tissues by decreasing the contents of the cytokine and the adhesion molecule, and changing the expressions of the cytokine , the adhesion molecule and then- mRNA.
