Purpose: Proliferative vitreoretinopathy (PVR) is commonly complicated to retinal detachment and is characterized by the migration and proliferation of various cells, and the development of contractile cellular membranes intravitreous or on either side of the detached neuroretina. The PVR membranes are composed of several types of cells surrounded by extracellular matrix(ECM). The purpose of present study is to investigate the pathogenesis of the early stage of PVR, by the means of observation of REP proliferation and embryonic fibronectin (FN) reappearance in the traumatic rabbit retinal detachment model with PVR development.Methods: 1. Pathology and immunohistochemistry with antibodies recognizing PCNA, FN and cellular FN were conducted. 2. In situ hybridization with RNA probe was also performed to investigate FN splice variant expression during PVR membrane formation.Results: 1. The rate of PVR successful formation is 93.3% (14 715).2. PCNA was expressed in RPE in the retinal detachment (RD) area with the peak at about 7th day post-trauma, but not expressed in the retinal non-detachment area.3. Total FN was expressed in the normal eye tissue, and increased post-trauma in the PVR membranes; while cFNwas expressed from 3rd day post-trauma but not expressed in the normal eye tissue.4. Total FN mRNA was detected in normal eye tissue, and increased post-tauma in wound surrounding tissue; however, embryonic FN mRNAs containing EIIIA and EIIIB were not detected in the normal eye tissue, but detected post-tauma in the wound surrounding tissue, specifically expressed in flbroblast, RPE and glial cells. Conclusions : 1. This traumatic rabbit retinal detachment model can be produced efficiently, and used to study mechanism of initial cellular proliferation in PVR.2. RD might play a key role in the initial stage of PVR formation.3. FN mRNA and protein expression increased rapidly post-trauma, especially embryonic FN reappearance post-trauma suggesting a functional role in the early stage of PVR formation.
