Objective: Using Caihushugansan as a media, to study the protective effect of the method of method of soothing the liver and regulating qi ( MSLRQ ) on gastric mucosa of the psychological stress rats and its mechanisms.Methods: (1) The models of psychological gastric erosion were induced by stimulating the 6-OHDA-treated rats to attack the experimental rats as social psychological stressor. (2) The praxiological change of the experimental rats was evaluated to confirm the close relationship between the gastric erosion and the social psychological stress and emotional change. (3) The histopathological and ultra-pathological change of gastric mucosa of the rats was evaluated to make sure that psychological stress could lead to gastric erosion and the protective effect of MSLRQ on gastric mucosa of the rats. (4) To study the effect of MSLRQ on the gastric mucosal barrier, the GMBF of the rats was measured by the hydrogen gas clearance method, the depth of mucus on the gastric mucosa was measured by PAS dyeing method, and the expression of EOF and EGFR in epithelial cells of the gastric mucosa was observed by immunohistochemistry dyeing and mRNA in situ hybridization. (5) To study the effect of MSLRQ on the post-receptor signal transduction, the content of cAMP in gastric mucosa was detected by radioimmunoassay system and the expression of the c-fos gene was observed by immunohistochemistry dyeing.Results: (1) MSLRQ could relieve the effect of psychological stress on the emotion and behavior of the rats. (2) Strong psychological stress could cause gastric erosion in rats. MSLRQ could protect the gastric mucosa and relieve the degree of erosion. (3) MSLRQ could relieve the decrease of GMBF, increase the depth of mucus on the gastric mucosa.ameliorate the continuance of the mucus. MSLRQ did not affect the expression of EOF in gastric mucosa. However, it could reinforce the expression of EGFR, increase the content of cAMP in gastric mucosa and inhibit the over-expression of c-fos gene.Conclusion: MSLRQ can adjust the capability of anti-psychological stress in rats, maintain the system of the mucosal barrier so as to protect gastric mucosa.
