Objective:To Observe the effect of Caihushugansan on functional dyspepsia through animal experiment,meanwhile the expression of gastrin and Somatostatin in Hypothalamus and Gastric antrum were determined to reveal the mechanisms of the method of soothing the liver and regulating qi(MSLRQ) on treating FD from the view point of brain-gut axis and brain-gut peptide.Methods:Methods:32 rats were randomly divided into normal group, model group,Chaihushugansan group and domperidone group.(each n=8).The animal model of FD was established by constantly squeezing their tails except normal group for 3wk,twice everyday,each time 30 min.From the first day,141%Chaihushugansan decoction and 0.1%domperidone solution were administered respectively to Chaihushugan san group and domperidone group by gavage(every time 1.5ml/100g) and mean while distilled water were administered respectively to normal group and model group,twice everyday.The animal's general situation and amount of food and water were observed daily,body weight were measured before and after the experiment.In the 22th day,Hypothalamus and Gastric antrum were taken for observing.The rate of intestinal propulsion and general situation of stomach were observed. Histological investigation of Hypothalamus and Gastric antrum by HE staining. The expression of gastrin and somatostatin in hypothalamus and gastric of rats by immunohistochemical were detected.Results:(1)Model group appeared margination,pad batch,decline in excitement,activity and fight,getting sluggish,blunted in responsiveness,loose and withered hair,more residual in stomach,and inflammatory cell infiltration. The above manifestation in Chaihushugansan group and domperidone group were not so obvious compared with model group.(2)The amount of food and water and weight growth in model group were less than those of normal group (both P<0.05~0.01),while the amount of food and water and weight growth in Chaihushugansan group and domperidone group were more than those of model group(both P<0.05~0.01).(3) The expression of GAS in the hypothalamus and gastric antrum of Model group were less than those of normal group(P<0.05~0.01),while the expression of SS in the hypothalamus and gastric antrum in Model group were significantly increased than those of normal group(P<0.01). The expression of GAS and SS in gastric antrum of Chaihushugansan group and domperidone group were increased and decreased respectively,and the differences were significant(P<0.05~0.01).There were no obvious difference about expression of GAS,SS in the hypothalamus between domperidone group and model group(both P>0.05).GAS expression in hypothalamus of Chaihushugansan group were increased than those of normal group but there was no obvious difference in SS expression in hypothalamus between Chaihushugansan group and model group(P>0.05).Conclusions:①The hypo-expression of GAS and the over- expre ssion of SS in Central and Peripheral may be one of its Pathophysiological mechanisms on functional dyspepsia.②The method of soothing the liver and regulating qi can increase GAS expression in Central and Peripheral and decrease SS expression in Peripheral gastric antrum,which may be one of its therapeutic mechanisms on functional dyspepsia.
