Si-Jun-Zi Decoction (SJZD) is a classic and representative Chinese herbal formulae with the function of tonifying qi and strengthening spleen for treating patient with syndrome of spleen-deficiency. Previous studies from others have demonstrated that SJZD can modulate the functions of digestive system and immune system, and the main manifestations of spleen-deficiency syndrome are disordered and weakened absorbing function, and changed immune function, these can be partly reversed by taking SJZD. The impairment of intestinal mucosal immunity, which is an important component both of digestive system and immune system, has a close relationship with spleen-deficiency, for both the pathogenesis and manifestation are similar in some disease such as ulcerative colitis, inflammatory bossl disease etc. Polysaccharide, an important bioactive substance, has immunomodulating activities both to systematic immune system and local immune system of intestinal mucosa. Many membrane molecules on the surface of mucosa epithelial cells are polysaccharides or glycosylation compounds. The four kinds of Chinese herbs, i.e. Radix Codonopsis, Rhizoma Astractylodis Macrocephalae, Poriuio and Radix Glycyrrhizae, formulating SJZD, are rich in polysaccharides. From the mentioned-above evidences and links, we could logically hypothesize that the total polysaccharides of SJZD (SJTPS) may play an important role in the exertion of bioactivity of SJZD, and may enhance the intestinal mucosal immunity, SJTPS can exert modulate function both in intestinal mucosa absorption and in immunity via the effect of intestinal epithelial cells. The effect of SJTPS and each single polysaccharides on rat intestinal epithelial cell line IEC-6 was investigated including the effect on cell proliferation, cell migration, cell adhesion and the change on the gene expression. The experiments showed that: 1.Effect of total polysaccharides of Si Jun Zi Decoction and each single polysaccharides onproliferation of IEC-6 cell line.It was indicated by MTT assay and cell count assay that purified compound in SJTP(PPS) which had been removed protein and polysaccharides which not removed protein(PS) could both promote IFC-6 cells growth and the effect ot PPS on the enhance the cells numbers an IFC-6 was better than that of PS. Continuing observation of incubating IEC-6 cells with SJTP demonstrated that PPS could accelerate IEC-6^ cells growth at different doses and in different time. However, most significant effect of PPS appeared when PPS incubated with IEC-6 cell line in dosage of 200 g/ml for 1 day and incubated for 2 days in dosage of 100g/ml. which indicated certain time-effect relationships was in existence in effect of PPS on impro\ement on IEC-6 cells growth. PPS could recover IEC-6 cells growth inhibition caused by Indomethacin. There were difference between the effect of PPS and each single polysaccharides on IEC-6 cells proliferation. The polysaccharides of Poriaio had obvious effect of enhancing cell growth, polysaccharides of Radix Clycyrrhi:ae and Radix Codonopsis made no effect, while polysaccharides of Rhiiomu Astractylodis Macrocephulae could inhibit cells growth markedly. The effect of all combination of any two polysaccharides on IEC-6 cell proliferation, namely pohsaccharides from Radix Codonopsis plus polysaccharides from Rhizoma Astractylodis Mucmcephalae, pohsaccharides from Radix Codonopsis plus polysaccharides from Poriaio. polysaccharides from Radix Codonopsis plus polysaccharides from Radix Glycyrrhizae, pohsaccharides from Rhiioma Astractyiodis Macrocephalae plus polysaccharides from Poriaio, polysaccharides from Rhizomci Astractylodis Macrocephalae plus polysaccharides from Radix Cilycyrrhizaz, polysaccharides from Poriaio plus polysaccharides from Radix Glycyrrhizae. were much weaker than PPS. This reflects that the complex proscription in traditional Chinese medicine is reasonable, and the function of Chinese herbs in a formulae is complementary to each other. 2.
