Background and Aims It is definite that the chronic hepatopathy is threastening health of human beings seriously,and more than one half of the infection of HBV occurs in China.Moreover,more than 80% of patients with primary hepatocellular carcinoma have positive HBsAg.In recent years,it has been found that helicobacter hepaticus(Hh) is etiological factor which could evoke chronic hepatopathy and lead to liver cancer in susceptible mice.Furthermore,nanobacteria have been found in human bile.However,it does not know the infectious condition of nanobacteria in Chinese healthy people and whether nanobacteria have relationship with human chronic hepatopathy and primary hepatocellular carcinoma.In this study,we investigated the infectious status of nanobacteria on serum of Chinese healthy people,and serum and liver tissue of patients with chronic hepatopathy and primary hepatocellular carcinoma.We expect the research could ensure whether nanobacteria lie in liver tissue of human beings,meanwhile it will give us a new knowledge to the development of chronic hepatopathy and primary hepatocellular carcinoma and to better prevent and treat liver cancer.Methods (1) We detected nanobacteria in serum of 336 cases of healthy adults in China by ELISA and immunohistochemistry stain to know the infectious status of nanobacteria in Chinese healthy people. (2) Resected tissues in cancer and beside cancer at least 2cm were cultured in 43 patients with primary hepatocellular carcinoma,while 15 liver samples of normal control were also cultured.Then we observed growth of the cultures and identified nanobacteria by several ways such as immunohistochemistry, immunofluorescence stain,calcific stain andTransmission Electron Microscopy(TEM). (3) Nanobacteria in the liver tissues were detected by immunohistochemistry to find the their infectious evidence in 28 chronic hepatitis B,25 posthepatitic cirrhosis,43 primary hepatocellular carcinoma and 15 normal control cases. Furthermore,part of tissues of cancer and beside cancer with positive immunohistochemistry were observed by TEM. (4) The nanobacteria in serum were detected by ELISA, immunohistochemistry stain and calcific stain in 25 chronic hepatitis B,30 posthepatitic cirrhosis,43 primary hepatocellular carcinoma cases, and the infectious rate were compared to that of healthy persons.Results (1) The infectious rate of nanobacteria in the healthy adults is 8.0% by ELISA,and 5.7% by immunohistochemistry stain.There is not difference between the two ways and between man and woman.Moreover,the age and sex could not increase the infectious risk of nanobacteria.However, the infectious rate is lower in those below 30-year-old than that of people over 60-year-old (p<0.05) . (2) The incidence of the white precipitation adhering to the culture tube is 34.9%,20.9% and 13.3% respectively ,by the naked eyes, in the cultures of liver tissues of liver cancer,beside cancer and the normal. Furthermore,by immunohistochemistry stain the infectious rate is 23.3%, 16.3% and 0% respectively.There is not significant difference among the 3 groups (p>0.05) ;The positive rate in the cultures with white precipitation adhering to the tube are 40.0% and 66.7% respectively in cancer and beside cancer tissues,and the different positive rate in beside cancer group shows significant difference (p<0.05 ) . (3) To analyse the relationship between nanobacteria and the clinical, pathological features, we find those with positive anti-HBc have lower nanobacteria infectious rate than those with negative anti-HBc (p<0.05 ). Meanwhile,to study the risk factors among the many clinic and pathologic features using Logistic analysis,we have not find any of them are candidate. (4) Observing the culture precipitation with positive immunohistochemistry stain by TEM,we found ellipse or round-shapes microorganism with diameter of400-500nm and black shell in all of the samples,those are nanobacteria.(5) The positive rate of nanobacteria infection in cultures from liver cancer,beside caner and normal liver by...
