[Objective] To investigate the protective effect of intra oxygen injection against hepatic ischemia/reperfusion injury and try to expiore the mechanism , so that could provide experimental basement for increasing the liver tolerance of ischemia, protecting liver from hepatic function failure.[Method] 48 rabbits were randomized devided into four groups with: A .sham surgery group, B. ischemia/reperfusion group, C. I/R+intra oxygen injection from peripheral vein, D. I/R +intra oxygen injection from hepatic artery. Set up hepatic ischemia/reperfusion model following Pringle's way .Compare four groups on the following parameters at 1 hour, 2 hour, 24 hour after surgery, the liver function (AST,ALT,LDH), the content of TNF- a in plasma, anti-oxidation capacity of the liver tissue(SOD , GSH-px , CAT) , energy metabolism(ATP, EC value), the content of MDA, testing of cell apoptosis , the expression of MMP-l/TIMP-1 in the liver tissue,histological change of the liver tissue and the survival rate of one week after surgery.[Result] 1. Compare to Group A, the injury of liver function of the Groups B, C, D were more serious, serum AST, ALT, LDH suspension increased(P<0.01 or P<0.05). The content of TNF-a in plasma were higher(P<0.01 or PO.05). The ATP content of liver tissue and EC value decreased (P<0.05). The content of MDA were higher (P<0.01 or PO.05). The cell apoptosis significantly decreased (PO.05); the positive rate of MMP-1 was significantly higher (PO.01 or PO.05), and that of TIMP-1 was opposite.The pathological change of liver tissue was evident (PO.05); The above indexes were molstly changed in Group B.There were some difference in the survival rate of one week after-operation, but had not significance statistically. 2.Compare to Group B, the injury of liver function of the Groups C> D were less seriously , serum AST, ALT, LDH suspension and the content of TNF- a in plasma were lower (PO.05); SOD,GSH-px, CAT of the liver tissue were more actively; the ATP content of liver tissue and EC value were higher (P<0.05), the content of MDA were lower (P<0.05); the cell apoptosis remarkably decreased (P<0.05); the positive rate of MMP-1 was significantly lower(P<0.05), and that of TIMP-1 was opposite; the pathological change of liver tissue were minor(P<0.05); the survival rate of one week post-operation were higher. 3. There were no significant difference between Groups C, D.[Conclusion] l.Intra oxygen injection can protect the liver from I/Rinjury, possibly by improve the energy metabolism of the ischemialiver tissue, decrease the generation of oxygen free radical and TNF ,the apoptosis of liver cells.2.There are no significant difference in giving intra oxygen injectionfrom the peripheral vein or the hepatic artery.3.MMP-1/TIMP-1 maybe take part in the procedure of I/R injury.MMP-1 possibly be a factor to induce the I/R injury.
