Experimental Study On The Therapeutic Mechanism Effect Of Gastric Carcinoma-Stopping Granule On Rats With The Syndrome Of Spleen Deficiency And Biood Stasis In Precancerous Lesions Of Gastric Carcinoma

In animal experiment, the models of precancerous lesions of gastric carcinoma were induced by MNNG and placing the spring in pylorus, which were in term with the syndrome of Spleen deficiency and Blood stasis in precancerous lesions of gastric carcinoma as to the projecting methods of the model,symptom changes and the level of the different objective mark ,for example substance P(SP),vascuactive intestinal peptid ( VIP ) ,6-ketone-prostaglandins F1a (6-K-PGF1a ), calcitonin gene-related peptide(CGRP), thromboxane B2 (TXB2),platelet alph-granule membrance protein 140(GMP-140), Epidermal growth factor receptor (EGFR),transforming growth factor alpha(TGF- a ) and vascular endothelial growth factor(VEGF). The rats was treated and protectively treated with Gastric carcinoma-stopping granule by means of Spleen invigorating and Blood promoting. The mucosal samples was taken form antral regions of stomach for pathological changes, immunohistochemicalstaining ,in-situ-hybridization and RT-PCT .The immunohistochemical staining was used to examine the expression of apoptisis. The level of the tumor suppressor gene P53 protein and the oncogene bcl-2protein were investigated by in-situ-hybridization. The relate of P53 and bcl-2 to the apoptosis was analyzed. The expression of TGF- a mRNA and VEGF mRNA were detected by RT-PCR. The Gastric carcinoma-stopping granule of Spleen invigorating and Blood promoting played an important role in gastric mucosal dysplasia, whether the Chinese herbs were used to treatment or protectively treatment . At first, the pathological changes of anthal regions of stomach was improved significantly. Secondly, the expression of SP, VIP,CGRP were raised, and intestinal peristalsis were strengthened. It reduced the lesions of the gastric mucosal . Trirdly, the level of TXB2 and GMP-140 in plasma membrance were decreased , but that of 6-K-PGF a were increased ,which activated the platelet. The apoptosis was raised by the gene of P53 and bcl-2 , whose expression was adjusted lowly . The secretory level of the cell gene of TGF- a , EGFR and VEGF were reduced , so as to stagnate the state of the cell proliferation ,at the same time ,alienate the cooperative effect of different factor in the process of precancerous lesions of gastric carcinoma.
