Experimental Study Of Tang Mai Ping In Treating Type 2 Diabetic Rats

Objective To investigate the effect of Tang mai ping (TMP) in treating type 2 diabetic and angiopathy of rats, and its pharmacological mechanism, to provide test data and theoretical direct for clinical treatment and new drug explorsion.Methods (1)Allocation of animal 45 of rats Mile wistar were divided into five groups by random sampling: normal control group (NC group), the Tang mai ping group(TMP group) the Yu quan wan group(YQW group), the Er jia shuang gua group (EJS group) and model control group(MC group). The control group was injected citrate buffer alone, the mode of type 2 diabetic rats was prepared by injecting low dosage (35 mg/kg) of streptozotocin(STZ) and feeding high sucrose and high fat diet. The TM group were ingested TM(2g/kg/d), YQ group were ingested YQ(0.45g/kg/d), EJ group were ingested EJ(0.15g/kg/d), the NC and MC groups were just ingested normal saline water. The course of treatment contin ued 10 weeks. (2)The body weight, the intake of chow and water of rats were checked. (3)We measured a serial of biochemical indeces. Such as serum glucose and LiPid. (4)The insulin free radical metablism and plasma endotheliolysin were defected with radio-immunicy. (5)Maximum binding of insulin receptor(1RMB) of RBC was observed, with radiolig and assay. (6) Immunohis-to-chemical study was used to detect the localization of insulin with microimage analysis system. (7)Is-lets morphology were observed with microscopy in HE staining. (8)The RNA expressing of Glucose transporter4 (GluT4) gene was detected with hyvirdism in situ.Results After treatment, the intake of chow of TM group was not decreased significantly than mormal control group(P>0.05), but was significant difference to MC group(P<0.01); the intake of water in TM group was significantly decreased than those in MP group(P<0.01), but was resmble to NC and YQ groups(P>0.05); the body weight of TM group was significantly decreased than that of MC and YQ group(P<0.01 orP>0.05), but was in- significant difference to NC and EJ groups(P>0.05). 2.Chang of biochemical indeces the fasting blood glucose(FBG) and 2 hours postprandial blood glucose(P2hBG) in TM, EJ and YQ groups were significantly decreased than that before treatment and MC group(P<0.01), and FBG in TM group was resemble to normal level(P>0.05), but in YQ group was still higher than NC group(P<0.05). The inpaired glucose tolerance was significantly high in TM group; Serum trig lyceride(TG) and total acholesterol(TC) in TM group were significantly decreased than those in MC group(P<0.05 and 0.01), the effect of TM decreasing the TG tevel was superior to that of EJ and YQ groups(P<0.05 and 0.01). 3 Chang of plasma endotheliolysin(ET) After treatment three treatment groups all sighificantly decreased ET level(P<0.01), MD group was significantly higher NC group(P<0.01), EJ level of was significantly decreased that of compared YQ and EJ groups(P>0.05). 4 Transformation of free radical metabolism. Serum LPO in MC group was higher than that of NC group(P<0.01), but SOD activity was lower(P<0.05), two treatment groups of Chinese medicine were significantly decreased Lop Level, and improved SOD activity(P<0.01), but those of EJ group were little different(P>0.05). 5 Chang of serum insulin (INS) and IRMB of RBC Serum INS in TM group was obviously lower, compared with Itself treatment(P<0.01), but higher compared with MC group and YQ group and was similar to that in EJ group(P>0.05); Serum IRMB of RBC in MC group was significantly decreased than that of NC group(P<0.01), Serum IRMB of RBC in three trentment troups were increased than that of MC group, and the differences were significant(P<0.01). 6 Iminunohistochemical study showed that the β cell experssion of insulin of TM group was increased comparead with MC group(P<0.01), the integral optical density of TM group was lower than that of control group(P<0.05), but the area percentage was resemble(P>0.05). 7 Islets morphology indicated that the secretive granule of islet - β cells were increased and islet & cells could be repaire...
