Effects Of Low Dose Androgen Treatment On The Heart Fuction In Male Rabbits Postinfarction Congestive Heart Failure Model

Objective: This study was designed to investigate the effects of low dose TU or DHEA on the heart function in rabbits postinfarction CHF model.Methods: 60 male New Zealand rabbits (4-5 month old ) were randomly assigned to ligation of first branch of left coronary artery(n = 52) or sham operation(sham group, n=8). After 3 weeks, all of the survived 40 MI rabbits were randomly treated with TU 3mg/kg/2weeks intramuscular injection (TU group, n = 10) , DHEA3mg/kg/d oral(DHEA group, n = 10), placebo (placebo group, n=10) for 3 months or were castrated (castration group, n = 10). Heart function were assessed by serial echocardiographic and in vivo hemodynamic parameters. Hormone changes were also recorded.Results: Serum FT and DHEAS levels in healthy male rabbits were 145.78 ± 15.42pg/ml and 0.078 ± 0.019 ug/ml respectively. Their levels decreased 30.5 ±16.9% and 30.4 ±13.9% at 3 weeks after MI. Serum FT level returned to normal ranges after low dose TU or DHEAtreatment, otherwise DHEAS level was higher than sham group. In placebo group, mean infarction area was 16.2 ±8.1%. Infarction resulted in enlarged weight corrected ventricular diastolic diameter and thickness of left ventricular wall, increased left ventricular end-diastolic pressure, decreased left ventricular ejection fraction and +dp/dtmax. Compared with placebo group, the weight of heart, left ventricular diastolic diameter and wall thickness of non infarction-area corrected by body weight decreased and LVEF increased in TU and DHEA group. Reversely castration decreased the body weight, enlarged left ventricular diastolic diameter and thicken the left ventricular wall. There was no improvement in LVEF.Conclusions: Serum FT and DHEAS levels decreased 3 weeks after MI in male rabbits. Low dose TU or DHEA treatment can remain normal serum FT level, improve ventricular performance and inhibit left ventricular remodeling in post infarction rabbit CHF model.
