Chronic aplastic anemia (CAA), with complex causings of disease and multiple pathogenic mechanism, is a refractory blood disease. Recent studies demonstrated that "soil hypothesis" ,one of the mechanism the attack of AA, closely related to matrix adhesion.In this study, the regulating function of Marrow-supplementing and Blood-engendering Granule (MSABEG) on adhesion function in CAA was explored by observing it's effects on cell adhesion molecules and cytokines interrelated to adhesion. To inquire into the mechanism of MSABEG, VLA-4/CD49d & c-kit/CD117 of bone marrow and TGF- 1 & IL-1 of serum in CAA patients were determined by using FASC and ELISA respectively before and after treatment by the Granule .The following results were observed:1) Curative effect of MSABEG is definite and prior to Aplastic Anemia Blood-engendering Tablet (AABET); 2) VLA-4/CD49d & IL-1 in CAA is in lowerly expressing state, and VLA-4/CD49d but not IL-1 can be significantly enhanced by MSABEG. The effect of the granule showed significant priority over Aplastic Anemia Blood-engendering Tablet; 3) c-kit/CD117 &TGF- 1 in CAA is in highly expressing state, but can be reversed by the granule.We conclude that abnormal function of adhesion exists in CAA, but Marrow-supplementing and Blood-engendering Granule, being proved improving adhesion function by regulating the level of cell adhesion molecules and interrelated cytokines at certain level, can prevent and cure CAA.
