Hypertension is a common cardiovascular disease. In about one third of cases with hypertension, itleads to left ventricular hypertrophy (LVH), which is one of main dangerous factors of human deathfrom cardiovascular disease, and an independent, primary factor and prognosticate index for suddendeath ,coronary heart disease and congestive heart failure(CHF). Though the mechanism is stillunknown, reversing LVH is the ultimate goal of treatment of hypertension and the research on itsmechanism becomes the hot point of studies in this field. With the development of pathophysiologicmechanism of hypertension, it is known to us that the essential of LVH lies in the injury of regulatorycapability of heart itself. Therefore, the treatment strategy is now transferred from amelioratinghemodynamics to protecting target organ and then improving life qualities.Object Chinese traditional medicine has been made a big progress in the treatment of hypertension andachieved fairly good effects by nourishing yin to reduce pathogenic fire and activating blood andremoving phlegm. But the mechanism of traditional Chinese medicine (TCM) still needs further studies.Zishuijianghuoyin(ZSJHY), which came from experiences of Prof GAO Erxin, possesses good effectson decreasing hypertension and ameliorating clinical symptoms. Previous studies indicated it couldprevent and cure 2K1C-RHR model of LVH in rats. On the basis of previous study, according to theview of integrity of TCM , the present study is to probe the characters of syndrome in LVH and theregularity of the syndrome changes further by observing the syndrome of model rats in macro view anddetecting the regularly index of micro view. To explore the mechanism effects of ZSJHY the presentstudy observed its effects not only on experimental hypertension LVH but also on apoptosis ofmyocardial cell and associated genetic regulation, which provides the experimental and theoreticalbases for its clinical application and drug development.Method In the present study, two-kidney-one-clamp(2K1C) LVH model in rats was built by pressingunilateral kidney artery of rats. Model rats were administered with ZSJHY 20,40 g kg-1 d-1 at oneweek after the model was established. Enalapril 20mg kg-1 d-1was treated as positive control drug. Byobserving symptoms and using experimental techniques of electrophysiology, pathology,immunohistochemistry and molecular biology, the dynamic changes of the characters of animal biology,hemodynamics, left ventricle mass index, apoptosis of myocardial cell and associated geneticexpression in rats at different time during LVH formation were observed. And the pathophysiologicalmechanism of hypertension LVH and its change of syndrome of TCM characters were then discussed.The evaluation of the syndrome in the model was carried out in the different periods in order to build arepeatable animal model integrated disease with syndrome. At the same time, it was also observed thatthe effects of ZSJHY on LVH in the different periods and its relative mechanism.Results 1 Establishment and evaluation of 2K1C hypertension LVH model 2K1C-RHR is a common model for hypertension in, which characters of hyperactivity of yang5 英文摘要due to yin deficiency discovered in past study and accompanied LVH, whereas the onset time of themodel was reported differently. The present study designed morphologic observation in differentperiods such as the 1st, the 4th , the 8th and the 12th weeks. The results showed: 1.1 About 80% rats came into hypertension, whose blood pressure was increased stably andrapidly. Cardiac muscle hypertrophy was found after 4 weeks. Left ventricle mass index in model ratswas remarkably higher than that in the sham rats. The difference was more significant at 8th an 12thweeks.
