The incidence rate of prostate cancer is increasing in China, in the west country (especially in the United States), prostate cancer is the most common malignant solid cancer in men and the second cause of death and it is said that it is becoming the leading cause of death recently. The biological behaviors of prostate cancer are very complicate, there are 4 kinds of biological behaviors: latent cancer (which have no harm to patients health and survival time and patients haven't any clinical symptom as well as, therefore no need to treat), Clinical cancer (which can invade, progress and metastasis soon and cause the death of patient, therefore patients must be given effective and reasonable therapy timely), accidental cancer and occult cancer in addition. Cells that have the appearances of prostate cancer can be found in the prostate gland of nearly half of all men over the age of 50, yet the lifetime risk of a man being diagnosed with clinically apparent prostate is only 11%, and the lifetime risk of dying of prostate cancer is only 3.6%. Thus, some 3 clinical prostate cancers develop for every 1 that will de proved lethal. So not only economical and mental press but also serious side effects (erectile dysfunction, premature ejaculate, osteoporosis, et al) may be caused by the excess therapy to latent prostate cancer. Therefore, the academe put forward a new opinion—the ultimate strategy for defeating prostate cancer calls for developing ways to distinguish the harmless (indolent) cancer from the potentially lethal ones. It is very important to find effective prognosis molecular markers to forecast the biology behaviors of prostate cancer for clinical doctors, otherwise a right therapeutic plan could not be designed unless they knew the biology behaviors of prostate cancer. There are a lot of research articles on prostate cancer, but up to date, no effective prognosis molecular marker that is pivot to prostate cancer treatment has been affirmed. We selected 6 kinds of gene that were very important to tumor according to literatures, Furthermore we designed this plan to study the correlations between 6 kinds of gene (P53, Bcl-2,Bax, E-cd ,Cat,AR) with prostate cancer stage, grade ,PSA value ,endocrine effect and survival time respectively , then to evaluate if they may be regard as effective prognosis molecular markers of prostate cancer .Endocrine androgen-ablation therapy (including castration, complete androgen block, anti-androgen drug et al) is a dominating method of prostate adenocarcinoma, Many cases of prostate cancer initially are androgen dependent, but the response to androgen-ablation therapy is transient, and after 12 to 24 months (mean 18 months), the majority of prostate cancers relapse to the status of androgen independence, resulting in death. Despite the availability of various therapeutic approaches none has provided a marked survival advantage for patients in the androgen-independent stage of prostate cancer. Novel therapies for androgen-independent prostate cancer, therefore, are urgently needed. We evaluated the feasibility to use Chinese tradition medicine Arsenic trioxide (As2O3 ) to treat prostate cancer, which has been shown to be effective in leukemia, and acute promyelocytic leukemia, both in vitro and in vivo. As model cell lines, we used androgen-independent prostate cancer cell line PC-3.This research included five parts, The former 4 parts clinical research were about the discussion of prognosis molecular markers of prostate cancer, the last part experimential research was about the effect of arsenic trioxide on androgen-independent prostate cancer cell line PC-3 growth.Part 1: The Expressions and Clinical Significances of P53 in Prostatic CarcinomaPurposes To investigate the expressions and clinical significances of p53 protein in prostatic carcinoma, and evaluate if it could be a prognosis molecular marker in prostate cancer. Methods Formalin-fixed, paraffin-embedded tissue sections from 45 cases of prostatic adenocarcinomas (Pca) and 10 cases benign p...
