Study On Working Mechanism Of Both Methods Of Warm And Purgation And Warm And Checking Diarrhea To Immunological Injuries Of Ulcerative Colitis

ObjectiveUlcerative colitis (UC) is an chronic and recured disease characterised byunspecific proctitis and colitis.Professor Tian Delu,my tutor who studies UCbased on the theory of Traditional Chinese Medicine and Modern Medicine,believes that UC is most similar to relapsed dysentery according to alteratingsyndromes and manifestations in episode period and remission period. And themechanism of UC can be summarized that deficiency in spleen is its root whilemoist heat is its end, and its local phathology is characterized by bloodstasis while inner ulcer is its phathologic manifestations. As for treatment,Kui Jie Decoction, a successful recipe acquired in clinic by Professor Tian,is often used in acute period while Shen Ling Baizhu Powder and such as ZhuoYu Decoctions of Wang Qingren are aplied in remission period. This method notonly can relieve symptoms and decrease relapse rate, but never produces badreaction in long term. Professor Wang Xinyue, who inherited from Professor Tian, believes thatdifferent therapy be used properly in different periods of UC. Mostly, themethod of warm and checking diarrhea and the warm and purgation method areusually used in clinic. However, checking and purgation are both conceptsopposed each other in TCM, and how to use both methods properly so as toimprove effect of therapy is emphasis of our research,which has been exploredwidely in clinic. So, the objective of this research is to explore the effective time on UCrat and possible mechanism of both recipes, the Warm Spleen Decoction whichis a typical recipe of method of warm and checking diarrhea and the ZhenrenYang Zang Decoction which is a typical one of warm and purgation method bycontrast and analysis on different stages of pathological process with centreof immune damage, so as to provide experimental evidence for timely therapywith Chinese Medicine.Methods The animal model of UC is made by TNBS/ethanol method, and generalbehaviors and local pathological change is observed in 1 week, 2 weeks6 温下法和温涩法对溃疡性结肠炎免疫损伤的作用机制研究and 4 weeks of animal model. (1) Comparison and analysis on immune status: T Lymphocyte Transformationwas determined by method of MTT, and circulating immunocomplex was determined bysedimentation and turbidimetry of carbowax. (2) Comparison and analysis on disequilibrium of Th1/ Th2: IFN-γand IL-4in serum was determined by ELISA method. (3) Comparison and analysis on content of cytokine induced inflammation:TNF-αand IL-8 in serum and colon was determined by ELISA method. (4) Comparison and analysis on expression of chemotaxis cytokine MIP-1αmRNA: determined by method of RT-PCR. (5) Comparison and analysis on anti-impairment agent response: NO in serumwas determined by method of nitrate reductase, and iNOS in colon was determinedmyeloperoxidase(MPO).(6)Evaluation on plerosis of colonic mucous membrane: Issue morpholog andcontent of Epidermal Growth Factor(EGF) was determined by ELISAM method.Results (1) The UC model was made successfully according ot observation on generalstatus pathomorphological change and determination on activity of MPO in colonissue,and the model period can be divided into aucte inflammation period from1 week to 2 weeks and chronic inflamation period as 4 weeks after rat model. (2) Warm Spleen Decoction not only can decrease activity of MPO, improvesymptoms and relieve phathological changes, while the activity of MPO was notdecreased in group of Zhenren Yang Zang Decoction, but higher than that ofmodel group,moveover,local phathological status and general symptoms had beenworsen. However, general status and local pathomorphological change can beimproved by Zhenren Yang Zang Decoction in chronic period.
