| Objective: To compare effects by acupuncturing on the hand jueyin meridians' Neiguan, the hand shaoyin meridians' Shenmen, DU meridians' Renzhong, the foot shaoyin meridians Zhaohai on MCAO Rats, and infulence of expression of apoptosis, IL-1 /3 and Caspase-lrnRNA both brain and heart at the same time, and further to explore the correlation between the brain and the heart and its scientfic implication at various levelsMethods: The changes of electrocardiosignal in different groups Rats were recorded respectively by electrocardiograph . CK, CK-MB, LDH. α-HBDH of myocardial enzymes were tested by enzyme reaction rate or immunodepression method .The levels of serum IL-1β were measured serially with two-layer antibody sangwich enzyme-linked immunosorbent assay (ELISA). Expression of apoptosis. Bax and Bcl-2 were detected by immunohistochemical technique. And IL-1 0 rnRNA and Caspase-1 rnRNA were carried out by RT-PCR.Results:l.The rate of abnormal electrocardiogram were higher'than 68% in cerebral ischemia rats, changes of the ECG ST, Similar cardiac infarction arrhythmia and elc were its main types. The levels of myocardial enzymes and serum IL-1 were significantly higher than those of the normal control ( P < 0.01 ) . Furthermore, There were positive correlations abcut expression of apoptosis and Bcl-2 between brain and heart (p < 0.01 , p < 0.05 ) . There were still positive correlations about expression of IL-1 P and Cas Dase-1 rnRNA between brain and heart (p<0.01) .2. When electroacupuncture(EA) in acupoints Iseiguan, Shenmen ,the rate of abnormal electrocardiogram were significantly lower than when EA in acupoints Renzhong, Zhaohai and model control groupC p < 0.05 or p < 0.01 ).The quantity of CK, CK-MB of myocardial enzymes and serum IL-1 P decreased in varying degrees when EA in acupoints Neiguan Shenmsn or Renzhong (p < 0.05 or p< 0.01 ) ,effects were superior to those of Zhaohai and model control group (p< 0.05 or p< 0.01 ) .3. After EA in acupoints Neiguan, Shenmen, expression of apoptosis and bax are inhibited, and bcl-2 expression enhanced, bcl-2/bax was up-regulating ,the difference was very significant (p < 0.05 or p < 0.01).4.Compared with Renzhong and Zhaohai, there were effects of down- regulating expression of IL-1 3 and Caspase-1 rnRNA by EA in acupoints Neiguan> Shenmen, the difference was very significant (p < 0.05 or p < 0.01).Conclusion:1. Cerebral Ischemia infulented the changes of electrocardiogram >myocardial enzymes and serum IL-1 . Apoptosis and Gene Expression of IL-1P and Caspase-1 between Brain and Heart were positive correlations , itprompt that Cerebral Ischemia lead to myocardial damage , apoptosis and ischemic inflammatory reaction involved in its development ,there is a closely pathologic association betweet heart and brain.2.Electroacupuncture on the Pericardium meridian of Hand-jueyin'Neiguan and the heart meridian of Hand-shaoyin' Shenmen can remarkably improve eletrocardio-activity, inhibite myocardial enzymes and regulate the leves of serum IL-1 P ,and simultaneously inhibited apoptosis and expression of IL-1 ?and Caspase-1 rnRNA in both Brain and Heart tissues, the therapeutic effect on cerebral ischemia by acupucturing meridians related to heart prompted that there would be some regular association between heart and brain with the hand jueyin meridian and the hand shaoyin meridian, and its inherent law and mechanism still remain to be inquired further.3. It is possibly part of the mechanism of Acupuncturing on the Pericardium meridian of Hand-jueyin'Neiguan or the heart meridian of Hand-shaoyin' Shenmen on MCAO Rats and protecting Cerebral Ischemia and secondary myocardial damage that Expression of Apoptosis and Gene Expression of EL-1P and Caspase-1 were inhibited between heart and brain.4. Acupuncturing on different meridians ' acupoints,have different degree therapeutic effects.
