As it is a very important topic about the prevention and treatment of cerebral ischemia in modern medical field, the author, under the direction of Dr. Zhang Jiawei, combined the understandings of Meridian Theory in Chinese medicine and the new outcome of modern neural biology to explore the objectivity of the therapeutic effect of acupuncture experimentally, in order know more clear about the mechanism of acupuncture therapy in the treatment of cerebral ischemic diseases and to found the scientific basis of acupuncture treating wind-stroke.Cerebral ischemic disease falls into the category of "Zhong Feng (wind-stroke)" of Chinese Medicine. In the Internal Classic, Zhong Feng was described differently according to its stages come forth, as Pu Ji (fall), Da Jue(giant faint) in the period of losing consciousness, and as Pian Ku(half weakness), Pian Feng(hemiplegia), Yin Fei(aphasia) in the period of body functions disturbing. And in the two thousands history of knowing this condition later on, it is proclaimed the theory of outer (environment) factors stroke, then, proceeding inner(biological and psychological) factors stroke, and at last both outer and inner stroke theory. Till nowadays, the cause of this disease is concluded to be due to pathogenic factors of six, respectively, Xu(weakness), Huo(evil of fire), Feng(evil of wind), Tan(evil of Phlegm), Qi(disturbance of function) and Xue (disturbance of body structure), all of which rooted from the Yin deficiencies of the disordered organs of Gan(liver), Shen(kidney) and modified by the factors of climate, natural environment, emotions, diet, overwork, age and sex. And acupuncture recorded in the Chinese medical books in thehistory typified the style of single point performance before Tang(one thousand years ago), and multi points performance after Tang. And in the Compendium of Acupuncture and Moxibustion composed in the period of Ming(more than five hundred years ago), there appeared a special chapter about the systemic therapeutic performance of acupuncture in treating windstroke. And time came to the nearer age to find the merge of western understandings into the system of Traditional Chinese Medicine. Therefore modern practitioners of TCM gradually come to the common route of applying Ren & Du Vessels as basics in the treatment of cerebral ischemic diseases, as TCM theory bears the viewpoint that the two vessels generalize the functioning all the other meridians that hold together to contribute to the functioning of the brain. As a result, it was considered by the author to be a great opportunity to further clarify the actual functioning of Ren & Du acupuncturing on the ischemic injured brain.Therefore, to observe the effect of electroacupuncture on synaptic formation and synaptophysin expression in rats with cerebral ischemic injury, the methods of ultrastructural analysis and immunohistochemistry were applied in an experiment. The experiment showed that ischemic injured brain can cause degeneration of the presynapse compositions, synapse decreases in number and density, and low expression of synaptophysin in hippocampus CA1 area. By the 5th week post operation, the increase of neonate synapses and increased expression of synaptophysin could be noticed in model group and treatment group. In the treatment group, the reconvery of synapse number and density were the most obvious which was 93.8% of those of the sham operation group, and which was significantly higher than those in the model group. Compared with sham operation group, COD value of synaptophysin upregulated to 70% in the model group, and rised to 93.3% in the treatment group, with COD value in the treatment group significantly ( P <0.01) higher than that in the model group. Thus, it was convinced that electroacupuncture had the function of improving synaptic regeneration and thus, making up the functioning of synapse re-building.To investigate the effect of electroacupuncture on synaptic transmission in Dentate Gyrus of cerebral ischemic injured rats, electrophysiological technique was applied torecord the long-term potential (LTP) in both basic synaptic transmission and 200Hz high-frequency stimulation (HFS) induced LTP of the dentate gyrus in rat with cerebral ischemic injury. It was found that no significant changes of magnitude of population spike(PS) occurred within 120 min in sham operation group, while magnitude of PS in cerebral ischemic injury group begun to descend by 60% ±7% at 10th min. The result showed significant difference statistically (P<0. 01) by comparison of the two groups. However, 30min after LTP induced by HFS, the magnitude of PS significantly increased and continued for about 180min in both the sham operation group and the cerebral ischemic injury group, while 40min after treatment with electroacupuncture (120min after HFS), magnitude of PS begun to increase by 171% ± 22% at 90th min in the electroacupuncture group. And the result was also of statistical significance. (P>0. 05). Therefore, it was concluded that electroacupuncture can enhance the basic synaptic transmission and the magnitude of LTP induced by HFS of the dentate gynus in cerebral ischemic injury in rats.To explore the effects of electroacupuncture at Ren meridian on astrocytes in the hippocampus of brain ischemia rats, single and double immunofluorescence stainings methods were applied. And it was found that in the 7 days group ,compared with the control group, the number of GFAP, Nse immunofluorescence positive cells and GFAP/Nse double-immunostainings of hippocampus in electroacupuncture at Ren meridian group was not significantly different (P>0.05). However, in the 14 and 28 days groups, it was significantly increased as compared with the control groups, the number of GFAP immunostainings was significantly decreased compared with the control groups (P<0.01). In the 28 days groups, the number of Nse immunostainings was significantly increased compared with the control groups (P<0.01). Thus, it was concluded that electroacupuncture at Ren Vessel adjusts the astrocytes in the hippocampus of brain ischemia rats effectively,controls their excessive proliferation and advances their multi-potential characteristic,which in favor of the restoration of neural cells.To explore the effects of electroacupuncture at Ren Vessel on differentiation and proliferation of neural stem cells in the subventricular zone of brain ischemia rats, we...
