Objectives To investigate the effects of microwave coagulation therapycombing with HAS in suppressing the growth of reinoculated H22 tumorand analyse the local immune response to evaluate the efficacy of thetreatments.Materials and Methods10~8 H22 cells were injected into the skin of KUNMING mice subcutaneously. The H22 nodules growing to a size of about 10 mm in diameter 10 days after the inoculation were used for the experiments. Mice burdened with H22 nodules were divided into six groups. Each group contained 60 mice (the number slightly varying due to accidental deaths of mice during experiments).Group A, B, C received local microwaval hyperthermia at 55-60°C for about 5 seconds .The day after hyperthermia treatment,Group B had HAS in oral way on ten successive days;Group C had injection of HAS in local area of microwaval hyperthermia on seven successive days.Group D only use HAS in oral way on ten successive days.Group E had injections of HAS alone into the H22 nodules on seven successive days.(HAS, a kind of superantigen, kindly provided by Xiehe company,Shenyang,China).Group F was the control group and received no treatment.At the same day,every group was been divided into three subgroups, and 10~7,10~6,10~5 H22 cells were injected into the skin of KUNMING mice in each subgroup respectively.The diameter of the reinoculated H22 nodules was measured once a week and surviving mice were counted every day .Tumor sizes were calculated from the length of the two axes (a,b;cm) of the nodules according to the formula [(a + b)/2]. The infiltration of immune cells(CD3,CD4,CD8) and cytokines(TGFβ,IL-2)into tumor and tissues adjacently was investigated and compared before andafter the treatments with HAS or/and microwaval hyperthermia. Results 1. The pathological features of subcutaneous tumor noduel were the same as that of ascites cancer cell of H22, which confirmed the success of inoculation.2. Complete coagulation necrosis and no cancer cell were observed histopathologically in the coagulated tissues the day after MTC.3. The general condition and the survival quality(1) No blood loss and quick recovery were found in group A, B, C.(2) The general condition of the mice in group A, B ,Cwas better than that in group D, E, F.(3) No local recurrence occurred in group A, B, C.(4) 2-month survival rate: The rates were 100% in group A,B,C , 61.5%,78.6%,92.3%in group DI,DII,DIII, 61.5%,76.9%,92.3% in group EI,EII,EIII, and 23.1%,42.9%,57.1% in group FI,FII,FIII respectively. 2-month survival rates in group A, B, C were significantly higher than that in group D, E, F (P<0.05).(5) The more reinoculated cancer cells, the more cancer incidences, especially in group D, E, F. The increase of cancer incidence is slight in group B, C. In group III (10~5), cancer incidences of reinoculated tumor in group A, B, C, D, E were significantly lower than that in group F. In group II(10~6),cancer incidences of reinoculated tumor in group A,B,C,G were significantly lower than that in group D,E,F (P<0.05 or P<0.01) . In group I (10~7), Cancer incidences in group B,C were significantly lower than that in group A ,G (P<0.05) , but all of them were significantly lower than that in group D,E,F (P<0.05 or P<0.01) ,cancer incidences in group F was the highest in all of groups.(6)There were no significantly differences in growth rate of reinoculated tumor between group I and group II.
