Studies On Effect Of Deer Fetus Preparations On Health Function Of Old Rats And Its Mechanisms

Deer fetus refers to fetus with its placenta taken from gestational deer or the young that hasn't eat milk after birth.Its pharmacodynamics function is said to be "beneficial to kidney and strong to masculine, nourish weak and grow marrow" in, and deer fetus is also used for tonic among the people.At present, deer fetus is usually made into medicinal extract paste in traditional method, but its curative effects vary greatly in clinical efficacy. In order to utilize deer fetus better, the study was conducted.According to modern Chinese medicine science and anti-ageing theories, the main material is deer fetus and with American ginseng , bee pollen and Radix Glycyrrhiza,et al. 16 prescriptions of deer fetus praparation were made on the basis of zhengjiao experiment design of 5 factors and 4 levels and ultra-micro shatter processing; To study the health function on old female rats, and select better prescriptions,pharmacodynamics experiment was carried on,2 better prescriptions being selected; To choose the best prescription and study the health function on old male rats, single factor randomly experiment design was conducted, compared with deer fetus pure power and human placenta; And its nutrition ingredient , chemical ingredient and toxicity are tested; It is concluded that:1.The main points to develop deer fetus preparations are that: the first is rinsing absolutely,the second is beating, the third is drying,the fouth is conventional shattering ,the fifth is ultra-micro shatter,the sixth is mixing with supplementary material even-distributed,the seventh is disinfecting.the last is pouring into capsule. The conventional drying method is simple, active and nutrient ingredient lost less; The ultra-micro shattering method made cells being broken, and maintain ingredient of preparation completely,safely and naturally.2. Research of deer fetus preparations on health function of old female rats are showed that:deer fetus preparations can improve immune function of old femal rats including index of thymus and spleen,increased by 20.59% and by 325%, content of GLOB in serum increased by 212%,and positive percent of T-lymphocyte ANAE is unsignificant; They can improve antioxidative effects of old female rats,including increasing serum SOD activity by 17.78%,decreasing the content of serum MDA by 60.40%; They can improve endocrine function of old female rats , the concentration of serum estradiol(E2) increasing by 257.199% significantly, but effect on the weight of ovary and womb being unsignificant.3. Research of preparations of deer fetus and placenta on health function in old male rats are showed that: deer fetus preparations can improve immune function of old male rats including index of thymus and spleen,content of GLOB in serum ,and engulf index andengulf rate of phagocyte increased extremely significantly,of which 2 latter are increased extremely significantly; They can restrain the MAO-B activity in brain and liver, decreasing the MAO-B activity by 67.74%and by 66.73%, decrease the contents of Lipofuscin in heart and brain of old male rats,reduce Lipofuscin deposit in tissue, prevent cell structure and function from the harm of Lipofuscin; They have the function similar to hormone, increasing rate of living sperm and decreasing rate of abnormal sperm of old male rats significantly, but the effect on the index of attached to sexual gland and the concentration of testosterone in serum being slight.4. The nutrient ingredient and chemical ingredient in deer fetus and its preparations are tested. It is showed that there are rich nutrient and chemical ingredient including protein, 18 amino acid, E2 and trace elements, and T is found out in deer placenta and its preparations.The content of protain in deer fetus powder is by 84.87%, EAAJ of human placenta, deer fetus powder and deer fetus preparation II are by 77.898%, 76.053% and 74.702% respectively. Those nutrient and chemical ingredients form the foundation of great health function of deer fetus and its preparations.5. The toxicity experiment showed that the process of making the preparations is suitable and the products are nonpoisonous, the LD50 to mice being more than 19200mg/kg, without sperm distortion in mice.6. To sum up the previous literature and the experiment results in this paper, the health function mechanism of deer fetus preparations is that metabolization (free radical, supplement trace element and et al), immune function and centre and endocrine function are regulated, thus strong body and aging slow down.The experiment showed that deer fetus preparations are nutrient health food which is safe and high effective, of which the deer fetus preparation II is the best, the prescription contains deer fetus power, Panax quinquefolium Linn., TCM C, TCM D and bee pollen, the proportion is 40:5:0.9: 2:16. The form of the deer fetus preparation may be capsule. The mechanism of health function is that antioxidative, immune and centre endocrine effects are regulated to maintain inside envirment constant, to increase the adaptability, to prevent disease and strong body.
