Abstract/Summary:
Cerebral infarction is one of the most serious diseases with higher incidence, mortality, mutilation rate, which brought huge economy and mental stresses for family and societies, It is an urgent duty for people to develop the available drugs to overcome the disease. It has been believed that there were two kinds of available strategies currently on acute cerebr- al infarction, ie: ① thrombolysis in superearlier time(within 3h in general, some delay it till 6h) ② neuroprotective treatments. In addition,there are other available methods such as anticoagulate,antiplatelets gathering, improve the blood circulation etc. though there are many kinds of ways and drugs on treatment of acute cerebral infarction , most of which only can improve some facet of complicated pathological changes. it is not only invalid but also can make condition worsen If these methods were taken inappropriately .owing to complicated pathophysiology process of cerebral infarction ,the modern medical science unanimously thinks that treatment to cerebral infarction can`t depend on single way but claim to comprehensive treatment with effect on many pathway and multiple link, namely so-called" the chicken tail wine" therapy, therefore a great deal of medicine have to be used at the same time necessarily, which enhance side effect of the medicine undoubtedly. However, the Chinese herbal medicine has an particular advantage at this aspect because of scientific compatibility and pharmaceutical technique, not only have no obvious side effect but also can attain multi- target treatment on acute cerebral infarction. HEEL is extracted from hirudo and earthworm by low temparature frozing-dissolved and membarence separation technology, Several studies found that the components of HEELsuch as hirudin from hirudo has anti- coagulant effect ,lumbrokinase from earthworm has positive effect on prompting tPA secretion and production. In addition, some studies shown that both earthworm and hirudo had effect on inhibiting inflammation and free radical injury. so we consider that HEEL should have above effects, and hold multi-target effect. It is essential that systemic and overall mechanism was investigated because it was kept unknown up to present.Objective To observe the theroputic effect of HEEL on rats with acute cerebral infarction. Methods A focal cerebral ischemia model of rats were established by embolizing left middle cerebral artery with autologous blood clots. The rats were divided into 3 group: Sham operation group ,model group and therapy group , therapy group was injected via tail vein with HEEL, both Sham operation group and model group were given normal saline , 0.25 mL.kg-1,once daily for 5 days. The changes of neurobehavior, infarction volume,brain water content, occlusion and recanalization of vessels, as well as histopathological changes were observed in 3 groups by neurological deficit evaluation ,TTC staining, dry-wet method , Ink-dextran infusion ,light microscope and electric microscope,respectively Results To compared with Sham operation group, the neurological deficit score(2.2±0.9), infarction volume(143±62),brain water content (84.4±3.2)in control group was remarkably increased, there was much less microvessels and sever histopathological changes. However, to comparedwith the control group, the neurological deficit scores(1.7±0.8), the infarction volumes (72±33), the brain water contents(79.7±3.7 ) in HEEL group were decreased, and histopathological changes were alleviated, microvessles were significantly increased, there is significantly difference between HEEL group and model group(P<0.01). Conclusion HEEL could improve neurological function, decrease infarct volume, abate brain edema and histopathological changes, has a significant theroputic effect on rats with acute cerebral infarction.Objective To investigate the mechanisms of HEEL on rats with cerebral infarction.
