In this paper,TCDCA anti-inflammatory mechanism, immunopharmacology and pharmacokinetics were studied.Studies on TCDCA Anti-inflammatory Mechanism Freund's complete adjuvant arthritis rats were used as a model of arthritis animals. TCDCA was intragastric administrated. The paws swelling of AA rats were measured in different inflamed periods. The contents of NO, LTB4, IL-1β, IL-6 and TNF-αof peripheral blood were measured by fluorospectrophotometry and ELISA methods respectively in AA rats. After TCDCA were successively intragastric administrated for 21d, the results showed that TCDCA markedly inhibited AA rats hind paws swelling in different inflamed periods,degraded the content of NO and enhanced the contents of LTB4, IL-1β, IL-6 and TNF-αin blood serum. After TCDCA were successively intragastric administrated for 4d, TCDCA degraded the content of LTB4 in peripheral blood remarkably. The anti-inflammatory action of TCDCA has relationship with inhibiting the production of LTB4 and NO.Studies on TCDCA immunopharmacologyThe thymic and splenic indexes were determined by weighting method respectively in control group of AA rats. The content of hemolysin in blood serum was measured by spectrophotometric method; the content of IgG in AA rats blood-serum was measured by immunoturbidimetry; the percentages of CD4+,CD8+ and CD19+ lymphocytes were assayed by flow cytometry; the ala auris swelling was determined by delayed allergy method; the phagocytosis of mononuclear phagocyte system was determined by using carbon particle clearance test; the contents of lysozyme and GM-CSF were measured by nephelometry and ELISA method respectively. The results revealed that TCDCA could inhibit the weights of immune organs of normal mice and AA rats obviously. TCDCA evidently enhanced the content of hemolysin in blood serum obviously; degraded the content of IgG in AA rats peripheral blood; enhanced the percentages of CD4+,CD19+ lymphocyte and the CD4+∕CD8+ ratios; inhibited delayed allergy of mice; reinforced the phagocytosis of mononuclear phagocyte system and enhanced the content of lysozyme in blood-serum of mice. After TCDCA wa successively intragastric administrated for 4d, the content of GM-CSF was enhanced in blood-serum of AA rats significantly. Accordingly, TCDCA could regulate immunological function of body.
