Objective: To prepare suitable praeparatum of kaempferiae aetherolea, and to discover the restraining mechanism of kaempferiae aetherolea by way of observing the effects on cell proliferation of gastric carcinoma and related molecules on orthotopic transplanted model and gastric carcinoma cell. Methods: 75 mice were divided into control group, NS group, CTX group, dissolvant group, kaempferiae aetherolea emulsion group randomly after the orthotopic implanted model bearing gastric carcinoma cell line MKN—45 built. The administrations began from the 3rd day after the surgery, which were as follows: the mice in kaempferiae aetherole emulsion group were given kaempferiae aetherole emulsion 0.5ml ip one day, six times a week; in NS group given isotonic saline 0.5ml ip one day, six times a week; in CTX group given CTX dilution (50mg.kg-1.d-1) 0.5ml ip one day, three times a week; in dissolvant group given emulsion dissolvant 0.5ml ip one day, six times a week; in control groups given no medicine. The experiment terminated on the 6th week, the tumors were weighted and the cell cycle number and shape were tested. We observed the effect in various regulating factors of tumor by gene array. COX-2, P53 was stained by immunohistochemistry and their m-RNA expressions were detected by RT-PCR. Every group cells cultured by different pharmaceutical blood serums before OD value was measured by MTT method and carcinoma cell colony number on lavor carang was detected. Besides, the protein of COX-2, P53 was observed through Western Blot. Results: We gained uniform and stable kaempferiae aetherole emulsion, which can inhibit the growth and proliferation of gastric carcinoma in vivo and in vitro, and reduce the expression of COX-2, P53 mRNA and encoding protein. Conclusion: The anti-cancer effect of kaempferiae aetherolea emulsion is achieved by regulating gastric carcinoma cell cycle, restraining the proliferation of gastric carcinoma cell at the G1 phase.
