Objective To study effects of recombinant human hormone (rhGH) on cellcycle of human gastric carcinoma xenograft in nude mice.Methods The nude mice of this experiment were divided into control group,rhGH group, cisplatin (DDP) group and DDP+rhGH group. After cultured humangastric carcinoma cells MKN45 were inoculated in right buttock of nude mice andcarcinoma xenograft model was formed, the drugs were continuously given to nudemice for 6 days. Then all animals were killed after the experiment was finished. Thetumor inhibitional rate was counted by measuring tumor volume. Cell cycle of humangastric cancer MKN45, cell proliferation index (PI) and DNA inhibitional rate wereanalyzed by flow cytometry (FCM). Then the organ/body weight ratio of liver, kidneyand spleen was analyzed.Results Cell cycle: After drugs was given the cells in S phase ofDDP+rhGHgroup and DDP group decreased obviously compared with ones of rhGH group andcontrol group (P<0.05). But there was no statistical difference among cells inG0-G1 phase, G2-M phase, PI and DNA inhibitional rate. The apoptosis cells in rhGHgroup increased remarkably (P<0.05). Tumor: The tumor obviously grew slowlyand the tumor inhibition rate increased in DDP group and DDP+rhGH group,compared with rhGH group ang control group(P<0.05). However, there was nostatistical difference between DDP group and DDP+rhGH group or between GHgroup and control group (P>0.05). Weight and organ/body weight ratio: Weights ofthe nude mice in DDP+rhGtt group were heavier than the mice in DDP group (P< 0.05), but them in DDP+rhGH group and DDP group were lighter than that in rhGHgroup and control group. There was also no statistical difference between rhGH groupand control group in weight comparison. The organ/body weight ratio of liver in rhGHgroup increased obviously comparing with the other group (P<0.05). Theorgan/body weight ratio of kidney in DDP group and DDP+rhGH group increasedremarkably comparing with control group or rhGH group (P<0.05). The organ/bodyweight ratio of spleen in DDP group and DDP+rhGH group decreased apparentlycomparing with control group or rhGH group (P<0.01).Conclusion In vivo rhGH dose not accelerate the multiplication of humangastric carcinoma cells.
