Objective To clarify promoter activity of Candida albicans yeast-hyphal regulational gene(HYR1), this study focused on fractional analysis of 1.8kb HYR1 gene promoter activity. Metheods Six chimeric genes were constructed in which various lengths of sequences between 1800bp upstream of the start of transcription of the HYR1 and 43bp downstream of this site were fused to LacZ reporter gene. These recombinant plasmids were transfected transiently to yeast EGY48. The putative promoters activities were observed and compared by means of LacZ measurement in transfected cells. Result The LacZ expression was observed only in construction driven by 600~200bp and 1800~1000bp fragment. Conclusion Some essential elements might exist in the region of 600~200bp and 1800~1000bp upstream of HYR1 gene, The high potential promoter sequence is of great significance to search for specific DNA-binding proteins in activated HYR1 gene.
